響應面優化黃皮葉中的活性因子超聲提取工藝及其抗氧化活性研究

[摘要]目的優化黃皮葉中活性因子總黃酮的超聲提取工藝條件,并測定其抗氧化活性。方法以乙醇體積分數、超聲時間及料液比為影響因子,以黃皮葉總黃酮得率為評價指標,在單因素試驗基礎上,采用響應面法優選黃皮葉總黃酮超聲提取工藝,并測定黃皮葉總黃酮體外清除1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-1-picryl hydrazyl,DPPH)自由基、羥自由基和總抗氧化的活性。結果黃皮葉總黃酮的最佳提取工藝條件為乙醇體積分數18%、超聲時間73 min、料液比31∶1(mL/g)。在此條件下,黃皮葉總黃酮得率達到(1.300±0.006)%。黃皮葉總黃酮得率大小的主次因素為乙醇體積分數>超聲時間>料液比,其中超聲時間與料液比因素之間的交互作用顯著。黃皮葉總黃酮體外清除DPPH和羥自由基的IC50值分別為0.282 mg/mL、1.152 mg/mL,用三價鐵還原抗氧化能力(ferric-reducing antioxidant power,FRAP)法測得1 mg/mL黃酮的FRAP值為533.3 μmol/L。結論該所選工藝合理、可行,可用于黃皮葉總黃酮的超聲提取;三種測定方法均說明黃皮葉總黃酮在一定濃度下有抗氧化活性。

Optimization of extraction technology from Clausena lansium leaves by response surface and their antioxidant activity study

[ABSTRACT]ObjectiveTo investigate the extraction of active factors of total flavonoid from Clausena lansium leaves by ultrasound and the antioxidant activity of flavonoid.MethodsCompounds in-Clausena lansium leaves were extracted by ultrasound under different levels of factors,such as alcohol con centration,ultrasonic time and ratio between solid and solvent.Total flavonoids in the compounds were tested.Extraction was optimized by response surface.The antioxidant activity against 1,1-diphenyl-1-picryl hydrazyl(DPPH),the antioxidant activity against hydroxyl radical,and the total antioxidant activity were investigated.ResultsThe optimum extraction conditions were 18%volume fraction of ethanol,73 minutes of ultrasonic time and 31∶1 of solid-to-solvent ratio(mL/g).Under these conditions,the yield of total flavonoid was(1.300±0.006)%.The order for the factors of the yield of total flavonoid were volume fraction of ethanol,ultrasonic time and solid-to-solvent ratio.The interaction between the ultrasonic time and solid-to-solvent ratio was significant.The IC50of the obtained total flavonoid against DPPH and the ion ofhydroxyl were 0.282 mg/mL and 1.152 mg/mL,respectively.The ferric-reducing antioxidant power(FRAP)value of 1 mg/mL flavonoid was 533.3 μmol/L.ConclusionOptimization of extraction technology byresponse surface is convenient and feasible.In some certain concentration of total flavonoids,all of the three methods have showed the antioxidant activity of total flavonoids extracted from the Clausena lansium leaves.

黃皮[Clausena lansium(Lour.)Skeels]是蕓香科黃皮屬植物,主要化學成分有香豆素類、卡巴唑和酰胺生物堿類、黃酮等。黃皮葉,性味屬辛、苦、平,可疏風解表,除痰行氣,有治療溫病身熱、咳嗽哮喘、氣脹腹痛、黃腫、瘧疾、小便不利、熱毒疥癩等功效,在民間常用于治療急性黃疸型肝炎、感冒、胃痛、疝氣、牛臌脹、哮喘、高血脂等病癥[1]。……
