miR-451與腫瘤的相關(guān)性

[摘要]miRNAs是一類參與個(gè)體發(fā)育、細(xì)胞增殖、分化和凋亡等生物學(xué)過(guò)程并與許多疾病如癌癥等的發(fā)生和發(fā)展密切相關(guān)的小分子非編碼RNA。miR-451在造血細(xì)胞分化成熟過(guò)程中起著關(guān)鍵性作用。近年來(lái)研究發(fā)現(xiàn)其在一些腫瘤如胃腸癌、乳腺癌、肺癌、膠質(zhì)瘤等中也起著重要的作用。本文主要對(duì)miR-451與腫瘤發(fā)生發(fā)展的相關(guān)研究進(jìn)展作一綜述。

[ABSTRACT] MicroRNAs are a group of small non-coding RNAs that participate in a wide range of biological processes include individual development, proliferation, differentiation, apoptosis and closely related to the occurrence and development of many diseases such as cancer. MiR-451 plays important roles in promoting erythroid differentiation and maturation and also in some tumors such as gastrointestinal cancer, breast cancer, lung cancer, and glioma. In this paper, the recent advances in the roles of miR-451 in tumor development are reviewed.

microRNA(miRNA)是真核細(xì)胞內(nèi)廣泛存在的一類約由20~22個(gè)堿基組成的小分子非編碼RNA。miRNA主要通過(guò)與靶mRNA的3′-UTR互補(bǔ)配對(duì),其參與個(gè)體發(fā)育、細(xì)胞增殖、分化和凋亡等生物學(xué)過(guò)程,并與許多疾病如癌癥等的發(fā)生發(fā)展密切相關(guān)。近年來(lái)miRNA以其對(duì)生命活動(dòng)獨(dú)特且重要的調(diào)控作用而備受關(guān)注。腫瘤是目前嚴(yán)重威脅人類生命健康的一類疾病,研究發(fā)現(xiàn)miRNA介導(dǎo)了腫瘤(如白血病、肺癌、膠質(zhì)瘤、乳腺癌、胃腸癌等)的形成、發(fā)生與發(fā)展等過(guò)程。miR-451最早是由Altuvia等在2005年由人腦垂體RNA中提取并命名,位于染色體17q11.2,與原癌基因ERBB2(v-evb-B2 avian erythroblastic leukemia viral oncogene homolog 2,ERBB2)區(qū)域相鄰[1]。miR-451在造血系統(tǒng),血液系統(tǒng)疾病等方面都有重要作用,并且在白血病、肺癌、膠質(zhì)瘤、乳腺癌等腫瘤組織中低表達(dá)。上調(diào)miR-451,可以增加腫瘤的凋亡率。
白血病是造血系統(tǒng)中常見(jiàn)的惡性腫瘤,在兒童和青壯年惡性腫瘤中發(fā)病率居首位,然而目前發(fā)病機(jī)制及病因還不清楚[2]。對(duì)40例兒童早前B急性淋巴細(xì)胞白血……
