





摘 要:為了建立姜黃標準湯劑的HPLC指紋圖譜,以來自四川、云南和廣西的15個批次姜黃飲片為原料制備姜黃標準湯劑凍干粉,通過優化樣品提取工藝和色譜條件,對姜黃標準湯劑的主要成分進行HPLC分析和化學模式識別分析。結果表明:15批姜黃標準湯劑的HPLC指紋圖譜共標定了13個共有峰,確認了11、12和13號峰分別為雙去甲氧基姜黃素、去甲氧基姜黃素和姜黃素;通過主成分分析篩選出了2個差異性成分(3號峰和4號峰),但目前還不能確定其所代表的物質;15批姜黃標準湯劑的相似度評價結果均大于0.9,整體可以聚為2大類,其中S5、S11和S14聚為一類,其余批次聚為一類。該HPLC指紋圖譜方法呈現良好的精密度、重復性和穩定性,可為姜黃標準湯劑及中藥配方顆粒產品質量控制提供參考和幫助。

Abstract: To establish the HPLC fingerprint of turmeric (Curcuma longa L.) standard decoction for its quality control, turmeric decoction pieces processed with 15 batches of Curcuma longa L. raw meterial from Sichuan, Yunnan and Guangxi were prepared as freeze-dried powder in laboratory for later extraction. Subsequently, the extraction process and chromatographic conditions were optimized. The obtained standard turmeric decoction samples were performed the principle component analysis by HPLC and chemical pattern recognition. The results showed that a total of common 13 peaks in the HPLC fingerprints of the 15 batches of turmeric standard decoction samples were marked; peaks 11, 12 and 13 were identified as bisdemethoxycurcumin, demethoxycurcumin and curcumin; two different components (peak 3 and peak 4) were screened out by the principal component analysis, but the substances they represented were still uncertain. The similarities of the 15 batches of turmeric standard decoction were evaluated greater than 0.9, and they could be divided into two categories, among which S5, S11 and S14 clustered into one category, and the remaining batches clustered into the other category. This HPLC fingerprinting method presents good precision, reproducibility and stability, which can provide reference and help for the quality control of turmeric standard decoction and traditional Chinese medicine formula granule products.

姜黃(Curcuma longa L.)是姜科姜黃屬多年生草本植物,具有破血行氣,通經止痛功效[1],臨床上用于治療經行不暢、頸椎病和肩周炎的肩臂酸痛、慢性膽囊炎和膽石癥等疾病。姜黃主要含揮發油類和姜黃素類成分[2],其中姜黃素具有抗氧化、抗炎、抗腫瘤、抗糖尿病等藥理作用[3]。標準湯劑作為中藥飲片與中藥配方顆粒的連接橋梁,用于衡量中藥配方顆粒與其對應的單味中藥飲片臨床湯劑藥效物質是否基本一致[4]。中藥湯劑和顆粒劑都不能通過其形態學特征對其質量進行控制,因而利用HPLC指紋圖譜技術對中藥湯劑進行質量控制已成趨勢[5]。……
