摘要 [目的]研究滸苔代謝產物的提取方法。[方法]以滸苔為研究對象,用不同的細胞破碎方式、溶劑提取體系、料液比等對均一樣本的代謝產物進行提取,氣相色譜-質譜法(GC-MS)鑒定代謝產物成分,利用主成分分析等方法,對不同的代謝提取體系進行評價。[結果]溶劑體系甲醇-三氯甲烷-水(7∶3∶3)和液氮處理-球磨破碎-超聲波的破壁方法提取并鑒定出81種代謝產物,該方法具有高提取產率、高重現(xiàn)性、易操作和速度快等優(yōu)點。[結論]溶劑體系甲醇-三氯甲烷-水(7∶3∶3)和液氮處理-球磨破碎-超聲波破壁是適合滸苔代謝物提取的方法,該方法可為滸苔及大型綠藻的代謝組學研究提供參考。

Metabolite Profiling of Seaweed Enteromorpha Using Gas Chromatography-Mass Spectrometry

Abstract [Objective] To study the extraction method of Enteromorpha metabolites. [Method] Enteromorpha as the research object, using different cell crushing methods, solvent extraction system, solid-liquid ratio, the homogeneous sample metabolites were extracted; the metabolites were identificated by GC-MS; using principal component analysis method, different metabolic extraction systems were evaluated. [Result] A total of 81 metabolites were extracted and identified by the solvent system of methanol∶chloroform∶water (7∶3∶3) and liquid nitrogen treatment-milling-ultrasonic breaking method, showing high extraction yield, high reproducibility, easy operation and fast and other characteristics. [Conclusion] It is a suitable method for extracting metabolites of Enteromorpha which can provide reference for metabonomics study of Enteromorpha and large green algae.

滸苔(Enteromorpha)亦稱苔條、苔菜,屬綠藻綱石莼科,為大型海洋綠藻,廣溫、廣鹽、耐干旱,在我國近海廣泛分布。在適合條件下容易出現(xiàn)爆發(fā)型增殖,給水產養(yǎng)殖造成嚴重困擾,其生態(tài)危害受到廣泛關注。同時滸苔又有較高的營養(yǎng)價值,滸苔“燒末吹鼻止衄血,湯浸搗敷手背腫痛”,具有軟堅散結、化痰消積、解毒消腫等作用;滸苔富含蛋白質、纖維素、碳水化合物和維生素等營養(yǎng)成分,在日韓等國家作為中高檔的佐料消費[1],且價格昂貴。
滸苔在近海中的綠潮爆發(fā)極具規(guī)律:每年4、5月份在山東、江蘇、浙江等局部海域爆發(fā),爆發(fā)期間對當?shù)厮a養(yǎng)殖造成極大損失[2],對近海環(huán)境也產生不良影響,而其他時間附近海域則較少出現(xiàn)。……
