溫州地區原發性肝細胞癌TP53基因249密碼子突變研究

[摘 要]目的 檢測溫州地區原發性肝細胞癌的TP53基因249密碼子突變情況,并探討其在肝細胞癌中所起的作用及其臨床意義。方法 收集石蠟包埋的原發性肝細胞癌組織120例,應用Sanger測序技術檢測突變情況。運用Spearman檢驗分析腫瘤組織中TP53基因249密碼子突變與原發性肝細胞癌患者臨床病理特征之間的相關性。結果 TP53基因249密碼子在120例肝癌組織中總突變率為3.33%(4/120),進一步分析顯示TP53基因249密碼子突變與肝細胞癌患者的臨床病理因素未發現存在相關性(P>0.05)。結論 在溫州地區,TP53基因249密碼子突變為非高發事件,未發現其與原發性肝細胞癌的進展相關。

Abstract ObjectiveTo investigate the mutation of TP53 gene codon 249 in primary hepatocellular carcinoma (HCC) in Wenzhou area, and explore its role in the development of HCC.Methods A total of 120 paraf■nembedded primary HCC tissues were collected, the Sanger sequencing technique were used to detect the mutation.The Spearman test was applied to analysis the correlation of the mutation of TP53 gene codon 249 and the clinicopathological features in HCC patients.ResultsMutation of codon 249 was found in 4 cases with a mutation rate of 3.33% (4/120); the research also showed that the mutation of codon 249 had no correlation with the clinicopathological feature in HCC patients (P>0.05).ConclusionOur data suggests that the incidence of mutation of p53 gene codon 249 in Wenzhou area is low and little contribution to the development of the hepatocelluar carinoma.

原發性肝細胞癌是最常見的惡性腫瘤之一,其發病機制是一個多因素、多步驟的復雜過程[1]。Bressac 等[2]發現肝癌中TP53存在基因的結構和表達異常,該基因發生突變后,抑癌功能喪失。后續研究報道[3-4],肝癌TP53基因存在多個位點基因錯義突變,其第七外顯子249位密碼子突變為為肝細胞肝癌所特有,在發病中起重要作用[5]。進一步研究[6-7]發現TP53基因249編碼子突變與黃曲霉素暴露密切相關。溫州市疾控中心腫瘤死因調查資料[8]顯示,溫州地區每10萬人中就有51人死于肝癌,而炎熱、潮濕的氣候,為該地區黃曲霉素的污染創造了有利條件。……
