目的 研究CD40配體基因rs7050168G/A位點(diǎn)多態(tài)性在廣西壯族及漢族人群中的分布，同時(shí)比較其在不同種族人群間以及同一種族不同性別之間，基因型和等位基因頻率分布的差異。

方法采用單堿基延伸PCR（PCR-SBE）的方法，分析201名廣西漢族人和199名廣西壯族人的CD40配體基因rs7050168G/A多態(tài)性。 結(jié)果 CD40配體基因rs7050168G/A位點(diǎn)表現(xiàn)出AA、AG和GG三種基因型，其在壯族人中的頻率分別為97.0%、1.5%和1.5%，而在漢族人中頻率分別為98.0%、1.0%和1.0%。CD40配體基因多態(tài)性在在廣西壯族和漢族人群之間比較差異都沒(méi)有顯著性（P均＞0.05）。進(jìn)一步將廣西漢族人群分型數(shù)據(jù)同人類(lèi)基因組計(jì)劃公布的4個(gè)人群相比，我們發(fā)現(xiàn)在廣西漢族人群中CD40配體基因rs7050168G/A位點(diǎn)基因型和等位基因頻率與日本和北京人群比較，差異均具有顯著性 （P均＜0.05），但與歐洲和非洲人群比較，差異沒(méi)有顯著性（P均＞0.05）。 結(jié)論 在廣西地區(qū)壯族及漢族人群中存在著CD40配體基因多態(tài)性，其基因多態(tài)性的分布頻率沒(méi)有顯著性的差異，但與其他種族人群比較存在著顯著差異。這種差異可能是導(dǎo)致一部分疾病在不同種族人群之間的臨床表現(xiàn)以及發(fā)病率存在顯著不同的因素之一。

［ABSTRACT］Objective To study gene polymorphism at rs7050168G/A of CD40 ligand in Guangxi Zhuang and Han populations，and to compare distributions of allele and genotype frequencies among different races and different gender in the same race.Methods The frequency of CD40 ligand gene rs7050168G/A allele and genotype were detected by using the method of polymerase chain reaction-single base extension （PCR-SBE）in 201 Guangxi Han and 199 Guangxi Zhuang populations.Results The genotype frequencies of AA，AG and GG of rs7050168G/A was 97.0%，1.5%and 1.5%in the Zhuangs，and 98.0%，1.0%and 1.0%in the Hans respectively.The frequencies of allele and genotype distribution of CD40 ligand geners7050168G/A polymorphism were not significant difference between the Zhuangs and the Hans （P＞0.05）.There were significant difference as compared with HapMap-JPT and HapMap-HCB （both P＜0.05），but compared with HapMap-CEU and HapMap-YRI，the CD40L polymorphism were not significantly difference （P＞0.05）.Conclusion There are CD40 ligand gene polymorphism in Guangxi Zhuang and Han populations.The frequencies of polymorphism distribution of CD40 ligand gene were not significant difference between the Zhuang and Han populations， whose distributions are significantly difference compared with others ethnic populations.The variation of CD40 ligand gene polymorphism among different ethnic groups might account for the varied clinical spectrum of some CD40 ligand related diseases.

CD40配體即CD40L，是TNF超家族的主要成員之一，表達(dá)于CD4+T細(xì)胞、柱狀細(xì)胞、血管內(nèi)皮細(xì)胞、單核細(xì)胞、自然殺傷細(xì)胞和血小板上，是一種分子量為39KD的Ⅱ型跨膜糖蛋白。CD40配體是體液和細(xì)胞免疫的中樞分子，與其受體CD40相互作用，作為T(mén)、B細(xì)胞活化的第二信號(hào)，介導(dǎo)T細(xì)胞依賴(lài)的B細(xì)胞活化和效應(yīng)T細(xì)胞產(chǎn)生，在體液免疫和細(xì)胞免疫調(diào)節(jié)中發(fā)揮著重要的作用。……
