【摘要】 脂聯素(Adiponectin,APN)是脂肪細胞分泌的一種細胞因子,在機體調節糖脂代謝、改善炎癥和免疫應答及抗動脈粥樣硬化等方面有著重要作用。以往研究多側重于脂聯素與糖尿病、心腦血管等方面的研究,而近些年關于脂聯素與抑郁癥之間的關系也逐漸受到重視,大量實驗研究已證明脂聯素具有抗抑郁的作用。本文回顧了脂聯素及其受體結構,并對新近發現的脂聯素與抑郁關系、及其相關發病機制等方面進行論述,為抑郁癥的治療提供提供新的研究方向。

【Abstract】 Adiponectin(APN) is an adipocyte-derived hormone that plays an important role in regulating metabolism of lipids and glucose,possessing anti-inflammatory and anti-atherogenic properties.Previous studies mostly focused on the relationship of adiponectin and diabetes or cardiovascular and other aspects,while the relationship between adiponectin and depression has been increasingly attracting emphasis in recent years.Emerging evidence suggests that adiponectin has an effect on anti-depression.This article is to review the structure of adiponectin and its receptor,and also to discuss the relationship between adiponectin and depression and the potential pathogenesis.And all of this will provide a new research direction for the treatment of depression.

隨著研究的深入,人們發現脂肪細胞分泌的細胞因子——脂聯素,不僅具有胰島素增敏、調節糖脂代謝等生物學作用,而最新研究顯示,脂聯素與神經精神疾病同樣存在密切關系。脂聯素可通過血腦屏障作用于腦組織,在調節神經興奮性及神經再生方面起到重要作用[1]。抑郁癥作為致自殺率最高的疾病之一,目前的藥物治療并不理想,因此新型抗抑郁機制有待發掘。近年報道發現脂聯素具有抗抑郁的作用[2],這為抑郁癥治療機制提供了新的前景。
1 脂聯素及脂聯素受體
1.1 脂聯素 脂聯素是由脂肪細胞分泌的一種可溶性防御性膠原樣血漿蛋白。人類脂聯素是由位于3q27的apM1基因編碼,約16 kb,包括3個外顯子和2個內含子[3]。脂聯素則由244個氨基酸組成,其分子量為30 kDa,蛋白結構包括了3個功能區:N端的信號肽區及種族特異性變異區、膠原重復序列和羧基末端的球狀結構域[4-5]。……
