摘要 [目的]研究用大孔樹脂吸附分離虎杖中白藜蘆醇的特性及其穩定性。[方法]分析大孔樹脂對白藜蘆醇的吸附曲線,分析流速、乙醇體積分數、提取時間、提取溫度、pH等因素對大孔樹脂吸附白藜蘆醇的影響。[結果]AB-8樹脂對虎杖中白藜蘆醇的吸附更接近單分子層吸附。以1.5 mL/min流速通過層析柱時吸附率更高,用70%乙醇提取虎杖中白藜蘆醇時提取率最高,3 h是最佳提取時間;60 ℃是最佳提取溫度;pH為4時白藜蘆醇提取率最高。穩定性研究表明,白藜蘆醇在酸性條件下保存濃度基本不變。[結論]用大孔樹脂吸附分離白藜蘆醇具有一定的優勢。

Research on the Adsorption Characteristics and Stability of Resveratrol from Polygonum cuspidatum

Abstract [Objective]The research aimed to study the adsorption characteristics and stability of resveratrol from Polygonum cuspidatum by macroporous resin adsorption.[Method]The adsorption curve of resveratrol was analyzed by macroporous resin.The effects of flow rate,ethanol volumn fraction,extraction time,extraction temperature and pH on the adsorption of resveratrol by macroporous resin were analyzed.[Result]The adsorption of AB-8 resin for resveratrol was more close to the monolayer adsorption.When the flow rate was 1.5 mL/min, the adsorption rate was higher; extraction rate of resveratrol from Polygonum cuspidatum with 70% ethanol was the highest;3 h was the optimum extraction time; 60 ℃ was the optimum extraction temperature; when the pH was 4, extraction rate of resveratrol was highest.Stability research showed that resveratrol concentration kept unchanged under acidic conditions.[Conclution]Adsorption and separation of resveratrol by macroporous resin has some advantages.

白藜蘆醇是植物受到外界刺激而產生的一種植物抗毒素[1-2],1940年首次從毛葉藜蘆根部分離得到[3-5],目前已在虎杖、葡萄、花生等至少72種植物中發現白藜蘆醇的存在[6-7]。研究表明,白藜蘆醇具有多種藥理活性[1-2],可預防癌癥的發生及發展,具有廣泛的應用價值,市場需求量大。白藜蘆醇的提取方法有微波輔助法、超聲提取法、超臨界CO2萃取法、酶法、有機溶劑提取法[8-10]。大孔樹脂是20世紀60年代末發展起來的一類新型樹脂[11],具有物理化學性質穩定、吸附和交換容量大、比表面積大、吸附速度快、選擇性好、吸附條件溫和、再生容易、強度好等優點,廣泛用于天然植物中各種活性成分的提取與純化[12]。筆者研究AB-8樹脂吸附葡萄和虎杖中白藜蘆醇的吸附特性、吸附的影響因素,尋找AB-8大孔樹脂吸附白藜蘆醇的最優條件,為更好地提取和分離白藜蘆醇提供一定的理論依據。……
