摘要 [目的]定位并比較不同日齡建昌黑山羊睪丸及附睪中GnRHR的分布及表達情況。[方法]分別收集55、104、300、1 140日齡建昌黑山羊的睪丸、附睪頭和附睪尾組織,然后進行脫水、包埋、切片后,采用免疫組化定位GnRHR在各個組織中的分布。[結果]GnRHR在建昌黑山羊睪丸、附睪頭和附睪尾組織細胞中均有分布,在上皮下層、基質層中分布較多,其表達量在300日齡的附睪尾和1 140日齡的睪丸中達到較高水平。[結論]GnRHR在睪丸、附睪中均有表達,而且不同日齡階段表達存在組織細胞特異性。

Localization and Distribution of GnRHR in the Testis and Epididymis from Different Age of Jianchang Black Goat

Abstract [Objective]To localize and compare the distribution and expression level of GnRHR in the testis and epididymis from different age of Jianchang black goat.[Method]The testis and epididymis were collected at 55,104,104,1 140 d after animal birth,then dehydrated,embedded and cut.Distribution of GnRHR was orientated by immunohistochemical assay.[Result]GnRHR in the testis,epididymal heads and tails,especially in subepithelial layer and stroma layer,in which had a higher expression level than other tissue was expressed.GnRHR expression was the most in the epididymis at 300 d and in the testis at 1 140 d.[Conclusion] GnRHR is expressed in the testis and epididymis,and its expression level has tissue-specificity at different age.

繁殖是生物物種延續最基本的生命活動,而動物繁殖必須依賴體內的繁殖調節物質。促性腺激素釋放激素(Gonadotropin Releasing Hormone,GnRH) 是下丘腦分泌的十肽,是下丘腦-垂體-性腺軸的關鍵調節因子[1],對于哺乳動物生殖器官發育和生理機能的維持起著關鍵的調節作用。GnRH作用的發揮依賴于促性腺激素釋放激素受體(Gonadotropin Releasing Hormone receptor,GnRHR) 的存在。GnRHR和GnRH 結合進而激發信號通路,并主要促使促黃體生成素 (LH) 和促卵泡素(FSH) 的合成與釋放,參與動物機體的繁殖調控[2-3]。
建昌黑山羊是四川省地方優良品種,廣泛分布于涼山彝族自治州。該山羊耐粗飼,易管理,抗病力強,繁殖力高,在當地地方經濟發展中占重要的地位[4]。目前有關雄性建昌黑山羊生殖生理的基礎研究很少。研究人員發現,抑制實驗動物和人的GnRH基因和GnRHR基因表達,會顯著抑制其生殖活動[5-6]。……
