摘要 [目的]分離并分析蘋果多酚組分及活性單體純度,并評價了它們的降脂活性。[方法] 利用乙醇提取蘋果多酚,并分離純化了4種(綠原酸、兒茶素、根皮苷、槲皮素)活性單體,采用高效液相色譜與質譜聯用分析多酚組分及活性單體純度,建立小鼠糖尿病模型。[結果]蘋果多酚中綠原酸的含量是35.207%、兒茶素3.991%、根皮苷15.662%、槲皮素0.274%。4個活性單體成分(綠原酸、兒茶素、根皮苷、槲皮素),經過液相質譜分析,綠原酸的純度是99.298%、兒茶素99.426%、根皮苷99.857%、槲皮素99.317%。動物試驗表明,蘋果多酚、綠原酸、兒茶素、根皮苷、槲皮素均能有效降低糖尿病鼠血總膽固醇(TG)、甘油三脂(TC)、低密度脂蛋白(LDL-c)水平,增加血高密度脂蛋白(HDL-c)水平。相比較蘋果多酚,4種單體(綠原酸、兒茶素、根皮苷、槲皮素)均顯示了更好的降脂活性。其中,根皮苷的降脂效果最好。[結論] 蘋果多酚及活性單體有著較好的降脂活性。

Extraction, Isolation and Purification of Apple Polyphenol and Its Active Monomers and Their Lowing-fat Effect

Abstract [Objective] To evaluate the lowing-fat effect of apple polyphenol and its active components. [Method] Apple polyphenol was extracted using ethanol, and four active monomers (chlorogenic acid, catechin, phlorizin, quercetin) were isolated and purified. High performance liquid chromatography coupled with mass spectrometry technology was used for analyzing components of apple polyphenol and purity of active monomers. A mice diabetes model was established. [Result] Apple polyphenol contained chlorogenic acid (35.207%), catechin (3991%), phlorizin (15.662%), and quercetin (0.274%). Purities of chlorogenic acid, catechin, phlorizin, and quercetin were respectively 99.298%, 99426%, 99.857%, and 99.317%. Animal experiments showed that apple polyphenol, chlorogenic acid, catechin, phlorizin, and quercetin could all effectively decrease blood TG, TC, LDL-c levels in diabetes mice, and increase HDL-c level. Compared with apple polyphenol, four active monomers (chlorogenic acid, catechin, phlorizin, quercetin) displayed stronger lowing blood fats effect. Among them, lowing blood fats effect of phlorizin was the best. [Conclusion] Apple polyphenol and its active components show good lowing-fat effect.

多酚為蘋果中的主要功能活性成分,蘋果多酚具有苯環并結合多個羥基化學結構,它主要包括黃酮醇、黃烷-3-醇、二氫查爾酮、羥基苯丙烯酸、花色素五大類。蘋果多酚純品為淡黃褐色粉末,略帶蘋果風味,稍有苦味,易溶于水和乙醇。蘋果多酚具有多種保健功能(例如抗腫瘤、防高血壓、抗突變、抗衰老、抗過敏、抗輻射等)[1],因此,它可以被使用在功能性食品的開發研究上,對人們的身體保健有著重要意義。……
