摘要[目的] 優(yōu)化魚(yú)體肌肉組織中脂質(zhì)過(guò)氧化物(LPO)含量的測(cè)定條件。[方法] 使用硫代巴比妥酸分光光度法測(cè)定LPO含量,設(shè)計(jì)乙酸用量、十二烷基磺酸鈉(SDS)用量、處理時(shí)間和溫度4因素3水平的正交試驗(yàn),考察各因素水平組合對(duì)魚(yú)體肌肉中LPO含量測(cè)定的影響,優(yōu)化測(cè)定條件。[結(jié)果]各因素對(duì)LPO含量測(cè)定結(jié)果的影響由大到小依次為SDS用量、乙酸用量、溫度、處理時(shí)間;通過(guò)直觀分析和方差分析,得出最優(yōu)組合為A3B1C3D2 ,即測(cè)定的最優(yōu)組合為20%的乙酸,65 ℃水浴中保持60 min。[結(jié)論]該研究可為魚(yú)類氧化應(yīng)激損傷研究提供方法支持。

Improvement of Thiobarbituric Acid Reaction for the Dertermination of Lipid Peroxidation in the Muscles of Fish

Abstract[Objective] To improve the optimum condition for measuring the lipid peroxidation (LPO) in the muscles of fish.[Method] The LPO was determined using spectrophotometer with thiobarbituric acid.The orthogonal design with four factors and three levels was conducted to evaluate the level of LPO in the cyprinoid fish.Four factors were included the gradient of acetic acid,sodium dodecyl sulfate,time and temperature,respectively.[Result] The weight sequence was known as SDS,the gradient of acetic acid,temperature,time by comparing the difference of each factor.The results indicated that the optimum group was A3B1C3D2 by visual analysis and variance analys,namely 20% acetic acid,65 ℃ water bath,60 min.[Conclusion] The study can support the research on the demage of oxidative stress in fish.

細(xì)胞在發(fā)生氧化應(yīng)激反應(yīng)時(shí),會(huì)發(fā)生脂質(zhì)過(guò)氧化,該反應(yīng)是發(fā)生于不飽和脂肪酸(UFA)共價(jià)鍵上的一系列自由基反應(yīng)。機(jī)體內(nèi)由于有很多UFA,因此極易受到氧化作用損傷,產(chǎn)生有細(xì)胞毒性的脂質(zhì)過(guò)氧化物(LPO)[1]。
目前,對(duì)脂質(zhì)過(guò)氧化的研究領(lǐng)域涵蓋了腫瘤、感染、衰老、心血管疾病、毒理學(xué)等方面[2-3]。測(cè)定脂質(zhì)過(guò)氧化的方法有熒光和分光光度法[4],由于分光光度法操作簡(jiǎn)單、快速,仍在大量使用,其中硫代巴比妥酸(TBARS)法是最為常用的方法[5]。一些脂肪酸,尤其是不飽和脂肪酸,氧化后逐漸分解為一系列復(fù)雜的化合物(如醛和酮類化合物),稱之為T(mén)BARS反應(yīng)物。通過(guò)檢測(cè)TBARS反應(yīng)物的含量來(lái)反映LPO的水平,從而表征氧化應(yīng)激反應(yīng)對(duì)細(xì)胞膜的損傷水平[6]。有關(guān)此方法優(yōu)化與改進(jìn)的報(bào)道較多[7-8],但鮮有專門(mén)針對(duì)魚(yú)體肌肉組織測(cè)定方法的報(bào)道,因此,有必要對(duì)其測(cè)定方法進(jìn)行優(yōu)化,以便適應(yīng)魚(yú)體肌肉組織中LPO含量的測(cè)定?!?br>
