摘要[目的]建立反相離子對液相色譜-二極管陣列檢測器測定紅薯粉條中溴酸鉀的分析方法。[方法]樣品以超純水超聲波提取,加入甲醇混合后離心,取上清液微孔濾膜過濾作為供試品液。使用Waters Symmetry shieldTM RP18色譜柱,以甲醇∶水(含4.0 mmol/L 檸檬酸與0.5 mmol/L十六烷基三甲基溴化銨溶液,pH為4.5)40∶60(V/V)為流動相,二極管陣列檢測器掃描檢測,以保留時間和190~400 nm光譜定性,210 nm波長下溴酸鉀峰面積定量。[結果]溴酸鉀在5.135~ 513.500 μg/mL濃度范圍內線性關系良好,相關系數大于0.999,方法回收率在83.5%~88.5%,檢出限為0.9 mg/kg,定量限為2.5 mg/kg。[結論]該方法簡單、快速、可靠、靈敏、高效,可用于紅薯粉條中溴酸鉀的分析檢測。

Determination of Potassium Bromate in Sweet Potato Vermicelli by Ionpair RPHPLC with Diode Array Detector

Abstract[Objective] To establish a method for determination of potassium bromate in sweet potato vermicelli by ionpair RPHPLC with diode array detector. [Method] The samples were extracted with ultra pure water by ultrasonic,mixed with methanol and centrifuged,supernatant solution was filtrated by microporous membrane as tested solution. The separation and determination were based on a Waters Symmetry shieldTM RP18 column with UV detector at 210 nm and methanol∶liquid (40∶60) of 4.0 mmol/L citric acid at 0.5 mmol/L hexadecyltrimethylammonium bromide(pH 4.5) as mobile phase. [Result] The calibration curve was linear in the range of 5.135 to 513.500 μg/mL and the correlation coefficients were greater than 0.999,while the recovery was from 83.5% to 88.5%. The limit of detection (LOD) and the limit of quantitation (LOQ) were 0.9 mg/kg and 2.5 mg/kg separately in this method. [Conclusion] The method is simple,rapid,reliable,sensitive and efficient,and can be used for the detection of potassium bromate in sweet potato vermicelli.

溴酸鉀是一種強氧化劑,能有效增強面團的彈性以及韌性,改善口感[1]。近年來的研究表明,溴酸鉀會損害人的中樞神經、血液及腎臟,國際癌癥研究機構也已將該化合物列為致癌物質[2-3]。我國衛生部在2005年明令禁止使用面粉處理劑溴酸鉀[4]。為降低成本,不法企業常以玉米淀粉等為原料偽制紅薯粉條,為了改善其品質,在加工過程中仍然存在違法添加溴酸鉀作為強筋劑的現象。
目前,溴酸鉀的檢測方法主要有分光光度法[5-6]、氣相色譜法[7] 、電化學分析法、液相色譜法和離子色譜法等[8-10],尚無紅薯粉條中溴酸鉀的檢測方法。現有溴酸鉀的檢測方法有一定的局限性,如分光光度法和電化學分析法存在專屬性不強的缺點;……
