[摘 要]目的探討建立以肝癌組織樣本為主的肝臟外科生物樣本庫及相關數據信息化管理的方法和標準化操作規程，為臨床與基礎研究提供高質量的臨床標本資源平臺。方法 收集肝臟腫瘤手術患者的血液標本（血清、血漿、白細胞）、組織標本(癌、癌旁、遠端正常組織)，將處理好的血液標本于-80 ℃冰箱保存，新鮮組織置液氮保存，定期隨機抽取液氮保存的腫瘤組織樣本進行RNA、DNA的提取及鑒定。結果現收集并保存有完整臨床信息及隨訪資料的肝臟腫瘤患者血液標本22 103份，組織標本3 319例，其中肝臟腫瘤組織標本807例，包括肝臟惡性腫瘤762例，良性腫瘤23例，肝硬化組織22例。隨機抽取組織RNA、DNA鑒定均符合標本質量標準，能夠滿足后續實驗要求。結論 初步建立標準化、規范化的肝臟腫瘤標本庫，制定了標準化的標本庫維護和管理規程，為肝臟腫瘤基礎和臨床研究提供了寶貴的實驗材料。

Abstract objectiveTo study the measures and standard operation procedure for establishing the biobank of liver surgical samples and information management of related data about samples,providing a high quality clinical specimen resource platform for clinical and basic research.MethodsBlood samples from patients with liver tumor(serum,plasma,leukocyte)and tissue samples(cancer tissues,tissues adjacent to carcinoma and the distal normal tissues)were collected,and then be kept in -80 ℃ refrigerator or in liquid nitrogen for preservation.Samples were randomly selected for the extraction and identification of RNA and DNA.ResultsA total of 22 103 blood samples with complete clinical information and follow-up data of liver tumor patients were collected and saved,807 cases of tissue specimens,of which there were 762 cases of malignant liver tumor,23 cases of benign tumors,22 cases of liver cirrhosis.Random tissue specimens of RNA and DNA identifications were preserved in high quality standards and could meet the requirements of the following studies.Conclusion A standardized and normalized liver tumor specimen biobank is established and the standard specimen library maintenance and management procedures are formulated,which provides valuable experimental materials for basic and clinical research for liver tumor.
