摘 要 采用ISSR分子標記技術對48份鳳凰單叢茶品種(系)進行了遺傳多樣性分析。選用10個多態性高、分辨力強的ISSR引物分別對供試材料基因組DNA進行擴增,共獲得121個位點,其中多態位點帶98個,多態位點比率(PPB)為81.0%。品種(系)之間的遺傳相似系數變化范圍在0.590 9~0.967 4之間。UPGMA聚類分析結果表明,在GS值為0.792的水平上供試材料可劃分為7大類,其親緣關系遠近與地理來源關系不大且與香味類型沒有必然的聯系。

Abstract ISSR molecular markers were used to analyze the genetic polymorphism of 48 Fenghuang-Dancong tea plant germplasms. Using 10 primers, we amplified 121 bands, of which 98 bands are polymorphic, and the average percentage of polymorphic bands was 81.0%. The genetic similarity among all the tested tea plant germplasms ranged from 0.590 9 to 0.967 4. Based on the genetic similarity and the UPGMA cluster, all the 48 accessions were clustered to 7 groups on the genetic similarity level of 0.792. The dengrogram indicated that the genetic of 48 tested tea plant germplasms had no certain relation with the geographical origin and aroma type.

鳳凰單叢茶屬烏龍茶類,產于“中國烏龍茶之鄉”的廣東省潮安縣鳳凰鎮,由茶農歷代沿習單株優選、單株培育、單株采制的傳統栽培方式和獨特的加工工藝制作而成的具有天然花香和特殊韻味品質的烏龍茶,現已形成了具有區域特色的品種(系)體系,享有“中國的國寶”之美譽[1]。茶業產量及品質提高依賴于茶樹優良品種的選育,而鳳凰單叢品種(系)資源繁多,且主要以香型為主線將其歸類,存在同一香型中包含多個品種(系),同型品種(系)間的品質參差不齊的現象,加之茶樹通常是通過雜交育種,從而使各品種(系)之間的親緣關系混亂,本底不清,遺傳背景不明確,極大地妨礙了品種(系)之間的親緣歸類、育種親本的選配和種質資源的創新和利用。
ISSR(Inter-Simple Sequence Repeat)是一種以基因組中常出現的SSR本身設計成錨定引物為單引物并基于PCR技術的簡單序列重復區間擴增多態性分子標記,具有無需預先知道基因組序列信息、快速、穩定、多態性豐富、重復性強及成本低等優點[2]?!?br>
