摘要[目的]應(yīng)用固相萃取(SPE)及高效液相色譜-串聯(lián)質(zhì)譜(HPLC-MS-MS)分析技術(shù),建立養(yǎng)殖水體中的18種磺胺類藥物殘留量測定方法。[方法]水樣中的磺胺類藥物通過HLB萃取小柱富集后,以C18反相柱為分析柱,0.1%甲酸水溶液和乙腈為流動相,采用HPLC-MS-MS進行定量分析。[結(jié)果]HPLC-MS-MS選擇多反應(yīng)監(jiān)測(MRM)正離子模式測定,可同時對養(yǎng)殖水體中的18種磺胺類藥物進行定性和定量。磺胺類藥物的檢出限均低于1.0 ng/mL,方法的平均回收率為70%~120%。[結(jié)論]該方法能夠簡便、快捷、有效地實現(xiàn)養(yǎng)殖水體中18種磺胺類藥物殘留的快速檢測。

Abstract[Objective] Using solid phase extraction (SPE) and HPLCMSMS analysis technology,determination method for 18 kinds of sulfa drugs residues in aquaculture water was established.[Method] Sulfa drugs in water sample were enriched through HLB extraction column,with C18 reversedphase column as analytical column,0.1% formic acid aqueous solution and acetonitrile as mobile phase,using HPLCMSMS to conduct quantitative analysis.[Result] Applying HPLCMSMS selective reaction monitoring (SRM) positive ion mode to determine,at the same time qualitative and quantitative analysis of 18 kinds of SAs in aquaculture water was conducted.The detection limits of SAs were less than 1.0 ng/mL,the range of the average recovery was 70%-120%.[Conclusion] The method is simple,quick and effective to realize rapid detection of 18 kinds of sulfa drugs residues in aquaculture water.

磺胺類藥物(SAs)是一種廣譜抗菌藥,在水產(chǎn)養(yǎng)殖行業(yè)中可作為飼料添加劑,且在動物疾病治療中也被廣泛應(yīng)用[1]。隨著我國水產(chǎn)養(yǎng)殖業(yè)的快速發(fā)展,磺胺類藥物的不合理使用,導致其通過動物糞便排入養(yǎng)殖水體的濃度增加。受污染的養(yǎng)殖水體匯入附近河流的地表水體后,會產(chǎn)生嚴重的環(huán)境污染及生態(tài)破壞問題[2]。因此,有必要建立養(yǎng)殖水體中磺胺類藥物的檢測方法,便于了解養(yǎng)殖水體及河流地表水體的污染狀況,及時采取有效的措施進行控制,降低環(huán)境的污染和生態(tài)的破壞程度。
目前,針對水體中的磺胺類藥物殘留的檢測方法有高效液相色譜法、酶聯(lián)免疫法等[3-4]。酶聯(lián)免疫法作為篩選方法得到了廣泛應(yīng)用,但容易出現(xiàn)假陽性;液相色譜法靈敏度低、選擇性差。……
