摘要:為優(yōu)化超聲波輔助提取劍葉龍血樹(shù)[Dracaena cochinchinensis (Lour.) S. C. Chen]樹(shù)葉中總黃酮的提取工藝,通過(guò)單因素試驗(yàn)和正交試驗(yàn),分析提取條件(提取劑乙醇體積分?jǐn)?shù)、料液比、超聲波功率、提取溫度、提取時(shí)間)對(duì)劍葉龍血樹(shù)樹(shù)葉中總黃酮得率的影響,確定劍葉龍血樹(shù)樹(shù)葉總黃酮的最佳提取工藝條件為超聲溫度70 ℃、提取劑乙醇體積分?jǐn)?shù)為70%、料液比1∶25(m∶V,g/mL)、超聲功率70 W,提取60 min。在最佳提取工藝條件下,劍葉龍血樹(shù)樹(shù)葉總黃酮平均率為1.605%。

Optimization of Ultrasound Wave Assisted Extraction Technology of Total Flavonoids from Leaves of Dracaena cochinchinensis (Lour.) S. C. Chen

Abstract:Explore the optimum extraction conditions of extracting total flavonoids from leaves of Dracaena cochinchinensis (Lour.) S. C. Chen by the assisted way of ultrasonic. To make sure the optimum extraction conditions, discussing the factors (ethanol concentration, solid-liquid ratio,the power of ultrasonic, extraction temperature,extraction time) affecting the yield through the single factor experiment and orthogonal experiment with ultrasonic assisted extraction. Optimum extraction conditions were 70 ℃ ultrasonic temperature,70% ethanol,1∶25 solid-liquid ratio(m∶V,g/mL), 70 W ultrasonic power and extracting 60 min. the extraction rate of total flavonoids from leaves of dracaena cochinchinensis could reach up to 1.605%.

劍葉龍血樹(shù)[Dracaena cochinchinensis (Lour.) S.C.Chen]在葉的基部和莖、枝受傷處常溢出少量的紅棕色液體,命名為樹(shù)脂,主要成分為血竭[1]。血竭具有活血散瘀、消腫止痛、止血等功效,起到抗菌、抗腫瘤、增強(qiáng)免疫等作用[2]。近年來(lái),中國(guó)對(duì)國(guó)產(chǎn)血竭的研究也逐漸發(fā)展起來(lái),血竭中的化學(xué)成分主要包括揮發(fā)油[3]、黃酮類(lèi)及酚類(lèi)化合物[4]。劍葉龍血樹(shù)根部及莖、樹(shù)葉均含有較多黃酮化合物,已有學(xué)者采用超聲提取[5]和微波提取[6],并取得比較好的提取效果。研究表明,黃酮化合物具有抗氧化、清除自由基的作用[7,8]。本試驗(yàn)以廣西劍葉龍血樹(shù)樹(shù)葉為原料,通過(guò)單因素試驗(yàn),結(jié)合正交試驗(yàn)探討超聲輔助提取總黃酮的工藝條件,以期為劍葉龍血樹(shù)的進(jìn)一步開(kāi)發(fā)利用提供依據(jù)。
1 ?材料與方法
1.1 ?材料與試劑
材料:劍葉龍血樹(shù)樹(shù)葉采自廣西崇左市江州區(qū)環(huán)城路,去掉劍葉龍血樹(shù)樹(shù)葉的兩端,洗凈晾干后切成小片繼續(xù)曬干,曬干后粉碎,過(guò)20目篩,備用;乙醇、亞硝酸鈉、硝酸鋁和氫氧化鈉均為分析純;蘆丁(批號(hào):21911,上海晶純?cè)噭┯邢薰荆!?br>
