摘要:用單因素試驗(yàn)方法探討超聲波輔助提取芙蓉花中總黃酮的最佳提取條件,考察了提取溫度、乙醇體積分?jǐn)?shù)、提取時(shí)間、超聲功率對(duì)提取結(jié)果的影響。結(jié)果表明,在提取溫度60 ℃,乙醇體積分?jǐn)?shù)80%,提取時(shí)間20 min,超聲功率400 W時(shí),芙蓉花中總黃酮的提取率為2.95%,提取效果最好,且優(yōu)于回流提取法。

Ultrasonic-assisted Extraction of Flavonoids from Flos Hibisci Mutabilis

Abstract: Single factor tests were adopted to optimize the ultrasonic assisted extraction of flavonids from flower of Hibiscus mutabilis (Flos Hibisci Mutabilis). The effects of extaction temperature, time, enthanol concentration and ultrasonic power on extraction ratio were studied. The results indicated that the best extraction efficiency could be achieved if extracting at 80 ℃ for 20 min when ethanol concentration was 80%; and ultrasonic power was 400 W. The yield of total flavonoids under these conditions was 2.95%; and it was superior to the heating recirculation method.

芙蓉花為錦葵科(Malvaceae)植物木芙蓉(Hibiscus mutabilis L.)的花,別名叫拒霜花,分布于四川、云南、山東等地。近年來(lái)研究證明,芙蓉的花、葉中含有蕓香甙等黃酮類化合物[1,2],因此芙蓉花可入藥,主治肺熱咳嗽、吐血、目赤腫痛等。但芙蓉花的藥用成分分析尚未見報(bào)道。黃酮類化合物是一類重要的天然有機(jī)化合物,廣泛存在于植物中,具有抗炎、抗菌、抗病毒、抗氧化、抗肝毒等多種生理活性,一直受到國(guó)內(nèi)外的廣泛重視,研究進(jìn)展很快[3-6]。為進(jìn)一步擴(kuò)展芙蓉花的藥用價(jià)值,對(duì)芙蓉花中總黃酮進(jìn)行了提取與測(cè)定。
1材料與方法
1.1材料
芙蓉花:采摘于成都市區(qū)。
儀器:722N分光光度計(jì)(上海精密科學(xué)儀器有限公司),KQ-400KDB型高功率數(shù)控超聲波清洗器(昆山市超聲儀器有限公司),AE240電子分析天平(瑞士梅特勒-托利多)。
試劑:無(wú)水乙醇、亞硝酸鈉、硝酸鋁、氫氧化鈉,均為分析純;蘆丁標(biāo)準(zhǔn)溶液(0.2 mg/mL)。
1.2方法
1.2.1樣品溶液的制備將芙蓉花洗凈后烘干,研磨成粉末后準(zhǔn)確稱取1.250 0 g粉末,加入乙醇溶液10 mL,經(jīng)提取,冷卻,過(guò)濾,從濾液中移取0.7 mL于50 mL容量瓶中用30%(V/V,下同)的乙醇定容。
