摘要:利用RT-PCR法對廣東煙草樣品的馬鈴薯Y病毒(Potato virus Y,PVY)輔助成分蛋白質(Helper component proteinase,HC-Pro)基因進行了克隆,并對其進行了序列分析。測序結果表明廣東PVY HC-Pro(命名為PVY-HC-GD)基因(用PVY-HC-GD表示)全長1 395 bp,編碼465個氨基酸。與已報道的PVY HC-Pro基因核苷酸序列相比發現,PVY-HC-GD與PVY NTN株系(PVYNTN)(AB331517)的PVY HC-Pro基因核苷酸序列相似性最高,達99.6%。而氨基酸序列比較表明,PVY-HC-GD與PVY N株系(PVYN)(AM268435)的PVY HC-Pro氨基酸序列相似性最高,為99.1%。進一步將PVY-HC-GD的核苷酸和其編碼的氨基酸序列與其他報道的同源基因比對構建了系統關系樹,結果顯示廣東煙草樣品中PVY-HC-GD與PVYNTN PVY HC-Pro親緣關系最近。

Cloning and Sequence Analysis of HC-Pro of PVY-GD Isolate

Abstract: Helper component proteinase(HC-Pro) gene was cloned by RT-PCR from tobacco leaves infected with Potato virus Y(PVY) in Guangdong, and then sequenced. The full length of HC-Pro of PVY from tobacco in Guangdong(PVY-HC-GD) was 1 395 bp, encoding 465 amino acids. Sequence analysis showed that PVY-HC-GD shared the highest nucleotide sequence similarity with Potato virus Y strain NTN (AB331517) (99.6%), and the highest amino acid sequence similarity with Potato virus Y strain N (AM268435) (99.1%). Phylogenetic trees constructed based on multiple alignment of nucleotide and amino acid sequences of PVY-HC-GD and 17 other PVY indicated that PVY-HC-GD had the closest relationship with Potato virus Y strain NTN.

馬鈴薯Y病毒(Potato virus Y,PVY)是植物病毒最大屬馬鈴薯Y病毒屬(Potyvirus)的典型成員。Potyvirus成員還與其他病毒間有協生作用,它通常作為激發病毒促進另一種異源病毒的復制,而其復制水平不變或稍有降低,如PVY就和馬鈴薯X病毒(PVX)之間存在協生作用,研究發現PVY促進PVX的侵染,使PVX的危害加重,而PVX對PVY則無影響,即PVY介導PVY/PVX的協生作用[1-4],病毒間的協生作用加劇了農作物病毒病的危害,造成了巨大的經濟損失。目前PVY在全球范圍內已成為馬鈴薯、煙草和辣椒等茄科作物病毒病的主要病源之一,并且在中國范圍內煙草遭受PVY危害呈逐年上升趨勢。
根據PVY的鑒別寄主反應、血清學反應和引起的癥狀類型將其分為多個株系,常見馬鈴薯Y病毒O株系(PVYO)、馬鈴薯Y病毒N株系(PVYN)和馬鈴薯Y病毒C株系(PVYC)三種株系。另外又有兩種株系,如馬鈴薯Y病毒NTN株系(PVYNTN)最早于20世紀80年代在匈牙利被發現[5],其與PVYN具有血清學關系,在煙草上引起葉脈壞死[6,7]。……
