色譜法測定藿香正氣水中乙醇含量*

用毛細管柱分離制劑中的乙醇,并以正丙醇為內標測定其含量。方法:色譜柱為毛細管柱zebron ZB-50(30m×0.32mm×0.25m);氮氣為載氣,柱前壓0.4MPa;分流比為30∶1;進樣口溫度為220℃,柱溫45℃;FID檢測器,溫度為250℃;進樣量1μL;正丙醇為內標物。結果:在該色譜條件下,藿香正氣水中乙醇和內標物得到良好的分離。范圍內線性良好(r=0.9995,n=7);平均回收率為97.0%。本法柱效高、方法簡單、分析快速、重現性好。

Abstract:The ethanol in medicament was separated by capillary column,and its content was determined with using n-propanol as the internal standard.The capillary column zebron ZB-50(30m×0.32mm×0.25m)was employed in this experiment,and the nitrogen and n-propanol was used as carrier gas and internal standard concentration,respectively.The pre-column pressure was 0.4MPa,the split ratio was 30∶1,the inlet temperature was 220℃,the column temperature was 45℃,the FID detector was used at 250℃ and the injection volume was 1μL.The results showed that under these conditions,the ethanol and internal standard in the Huoxiang Zhengqi solution was well separated.The linear relation was good in the range (r=0.9995,n=7),the average recovery rate was 97.0%.This chromatography method was high column efficiency,simple and fast and reproducible.

藿香正氣水是常用中成藥,主要作用是解表祛暑,化濕和中。適用于中暑,腹痛腹瀉,發熱等疾病[1]。藿香正氣水中乙醇含量是檢測的一項重要指標,2005年中國藥典一部中采用填充柱氣相色譜法測定乙醇含量[2]。本文采用氫火焰離子化檢測器-氣相色譜法測定藿香正氣水中乙醇的含量,為乙醇含量的測定提供參考。
GC-3900氣相色譜儀,FID檢測器,聚乙二醇毛細管柱zebron ZB-50(30m×0.32mm×0.25m)。甲醇、無水乙醇、正丙醇均為分析純,水為超純水。藿香正氣水,四川蜀中制藥有限公司。
2.1 色譜條件
聚乙二醇毛細管柱zebron ZB-50(30m×0.32mm×0.25m)柱溫45℃;進樣口溫度220℃;檢測器溫度250℃;靈敏度102;
載氣:純度為99.99%的高純氮氣,柱前壓0.4 MPa;
燃燒氣:純度為99.9%的氫氣,柱前壓0.4 MPa;
助燃氣:空氣,柱前壓0.4 MPa;
進樣形式:分流進樣,進樣量1μL,分流比為30∶1;
程序升溫:初始溫度:45℃,以5℃/min升至80℃。
色譜柱的老化:在通載氣的情況下,使柱溫升至160℃,保持 2~4h。……
