[摘要] 目的 觀察中介素1-53對大鼠肺缺血再灌注損傷(LIRI)肺組織過氧化物酶增殖物激活受體γ(PPAR-γ)的影響。方法 將健康Wistar 大鼠54 只隨機分為手術對照組(C組)、缺血再灌注組(IR組)、中介素干預組(D組)。每組分別在缺血45min、再灌注60min、120 min 3個時點處死6 只大鼠,觀察肺組織病理形態變化,觀察各組肺濕干質量比值(W/D)、CINC-1含量、PPAR-γmRNA、PPAR-γ蛋白的改變。結果 與IR組比較,中介素1-53預處理能夠明顯升高PPAR- γmRNA(P<0.05),減輕肺水腫,并降低CINC-1蛋白含量(P<0.05)。結論 中介素1-53能上調PPAR-γ的表達,可能通過PPAR-γ的抗炎作用對LIRI起保護作用。

Effects of Intermedin1-53 on Pulmonary Peroxisome Proliferator-activated Receptor-γ Expression in Rat Lung Ischemia-reperfusion Injury

[Abstract] Objective To investigate the effect of intremedin 1-53 on peroxisome proliferator-activated receptor-γ(PPAR-γ) in rat lung ischemia-reperfusion injury. Methods Fifty-four healthy Wistar rats were randomly divided into three groups:surgical control group(C group), ischemia-reperfusion group(IR group)and intermedin-pretreated group(D group). At three time points(45 min ischemia,60 min and 120 min reperfusion),6 rats from each group were killed,the lung histopathological changes were observed and the wet /dry weight ratio of lung tissue,CINC-1 content,PPAR-γmRNA and PPAR-γprotein in each group were determined. Results Compared with IR group,the pre-treatment by intermedin1-53 significantly increased PPAR-γmRNA(P<0.05),and decreased interstital edema and CINC-1content(P<0.05). Conclusion Intermedin1-53 may upregulate PPAR-γexpression,which has protective effects on rat lung ischemia- reperfusion injury.

中介素(IMD)是2004年由Roh等[1]用種系特征方法分析基因庫,從人類和其他脊椎動物基因組證實的一種新的降鈣素(CT)/降鈣素基因相關肽(CGRP)家族肽.IMD前體蛋白可被內源性肽酶降解為prepro-IMD95-147即IMD1-53,目前國內外有報道中介素1-53對油酸致急性肺損傷和心肌缺血再灌注損傷以及腎臟缺血再灌注損傷有保護作用[2-4],但中介素1-53對肺缺血再灌注損傷的研究尚未見報道。過氧化物酶體增殖物激活受體γ(PPAR-γ)屬于細胞核激素受體超家族的成員,在抗炎和免疫調節等方面有重要作用。本課題擬通過建立大鼠在體肺缺血再灌注模型,觀察肺組織PPAR-γ表達情況,旨在探討外源性人工合成的IMD1-53對肺缺血再灌注損傷的作用及其可能機制。
1材料與方法
1.1材料
1.1.1實驗動物健康雄性Wistar大鼠54只(山西醫科大學動物中心提供),體質量220~300g。
