





摘要 [目的]以絲瓜(Luffa ssp.)品種“徐綠一號(hào)”的未授粉子房為試驗(yàn)材料,分析不同因素對(duì)類胚體形成的影響。[方法]設(shè)置L16(215)正交試驗(yàn)分析子房發(fā)育階段、4 ℃預(yù)冷處理、35 ℃黑暗熱激處理、TDZ、6-BA、ABA、NAA、2,4-D對(duì)絲瓜離體子房培養(yǎng)獲得類胚體的影響。[結(jié)果]不同因素對(duì)子房片的發(fā)育過程具有不同的效果,其中采集當(dāng)天開放的雌花、4 ℃預(yù)冷處理、在MS培養(yǎng)基中添加0.04 mg/L TDZ對(duì)子房片的類胚體形成具有促進(jìn)作用,添加0.04 mg/L 2,4-D對(duì)絲瓜子房類胚體形成具有抑制作用,但可以促進(jìn)子房片生根;另外預(yù)冷處理與TDZ、TDZ與雌花采集時(shí)間、4 ℃預(yù)冷和ABA的兩兩交互作用對(duì)絲瓜類胚體的形成也具有顯著影響。[結(jié)論]有效形成絲瓜子房類胚體途徑:當(dāng)天采集子房+4 ℃預(yù)冷處理+0.04 mg/L TDZ。

Induction Study on the Pathway of Embryoid Formation in Ovary Culture of Luffa ssp.

Abstract [Objective]To analyze the effects of different factors on embryoid formation using unpollinated ovaries of Luffa ssp. cultivar ‘Xulv No.1’ as experimental material. [Method] An L16(215) orthogonal test was set up to analyze the effects of ovary collection time, 4 ℃ cold pretreatment, 35 ℃ dark heat shock treatment, TDZ hormone, 6-BA hormone, ABA hormone, NAA hormone and 2,4-D hormone on embryoid formation in vitro cultured Luffa ssp. ovaries. [Result]Different factors had different effects on the development process of ovary slices. Among them, collecting female flowers that opened on the same day and adding TDZ hormone to MS medium had a promoting effect on embryoid formation in ovary slices. However, 2,4-D hormone had an inhibitory effect on embryoid formation in Luffa ssp. ovaries but could promote rooting of ovary slices. In addition, the interaction between 4 ℃ cold pretreatment and TDZ hormone and between TDZ hormone and female flower collection time also had a significant effect on embryoid formation in Luffa ssp.. [Conclusion]The effective way to form embryoids from Luffa ssp. ovaries is: collecting ovaries on day of flowering+4 ℃ cold pretreatment+0.04 mg/L TDZ hormone induction in vitro.

絲瓜(Luffa aegyptiaca Miller)為葫蘆科(Cucurbitaceae)絲瓜屬(Luffa Mill.)一年生草本植物,其嫩瓜含有豐富的蛋白質(zhì)、碳水化合物、氨基酸等物質(zhì),具有保健和美容的作用[1];老瓜或藤蔓可用藥,亦可加工,具有特殊的藥用和工業(yè)價(jià)值[2]。絲瓜在我國(guó)南北均有種植,主要栽培種有普通絲瓜和有棱絲瓜2個(gè)類型[3]。近年來,絲瓜市場(chǎng)需求量不斷增大,對(duì)絲瓜不同品種的需求也在擴(kuò)大。
