





摘要 探討大蒜愈傷組織誘導及增殖分化的影響因素,建立大蒜愈傷組織誘導、增殖及器官分化的培養體系。結果表明,適合大蒜愈傷組織誘導及增殖的培養基激素濃度及pH為2 mg/L NAA,2 mg/L 6-BA,1 mg/L 2,4-D和pH 5.8;蒼山蒜愈傷組織出芽分化的最佳培養基激素濃度及pH為1 mg/L NAA,3 mg/L 6-BA,pH 5.8;蘇聯蒜愈傷組織出芽分化的最佳培養基激素濃度及pH為2 mg/L NAA,2 mg/L 6-BA,pH 6.1;蒼山蒜和蘇聯蒜愈傷組織生根分化的最佳培養基激素濃度及pH均為1 mg/L NAA,pH 5.8,表明高濃度的2,4-D有利于大蒜愈傷組織的誘導,但并不利于其分化。

Study on the Induction of Garlic Callus by Hormones and pH

Abstract The factors that affected callus induction, multiplication and organ differentiation of garlic were explored and the high efficient regeneration system was established. The hormone concentration and pH value of the culture medium suitable for garlic callus induction and proliferation were selected: 2 mg/L NAA,2 mg/L 6-BA,1 mg/L 2,4-D and the pH of the medium was 5.8. The hormone concentration and pH value of the culture medium suitable for garlic callus organ differentiation were selected. The optimal medium hormone concentration and pH value for callus differentiation of Cangshan garlic were 1 mg/L NAA,3 mg/L 6-BA and the pH of the medium was 5.8. The optimal medium concentration and pH value for callus differentiation of Soviet garlic were 2 mg/L NAA,2 mg/L 6-BA and the pH of the medium was 6.1. The optimal medium concentration and pH value for callus rooting and differentiation of two varieties are 1 mg/L NAA and pH 5.8. Research has also shown that high concentrations of 2,4-D were beneficial for the induction of garlic callus, but not for its differentiation.

大蒜引入我國馴化栽培已有2 000多年的歷史,具有較高的營養價值和經濟價值,尤其是大蒜素(蒜氨酸)經蒜氨酸酶作用形成的揮發性硫化物,使大蒜具有殺菌、抗癌、抗衰老等作用和獨特的大蒜風味。大蒜適應能力較強,在我國各地都有栽培,包括山東萊蕪、金鄉、蒼山,江蘇射陽、太倉,安徽亳州、來安等。我國是世界上最大的大蒜生產國和消費國,也是出口創匯大國,總栽培面積30萬hm2,干蒜頭總產量500余萬t,約占全世界總產量的2/3。
大蒜植株再生有2條主要途徑,即愈傷組織途徑和不定芽途徑,外植體的取材部位不同,誘導產生愈傷組織的能力也不相同,產生的愈傷組織在質地、形態、顏色、細胞組成及分化能力上也有明顯差異。……
