摘要[目的]分析動物雙歧桿菌AD011暴露外表面蛋白的種類和功能,有助于了解該菌株的各項生理功能以及為探討該菌與宿主相互作用的分子機制奠定基礎。[方法]以動物雙歧桿菌AD011基因組序列為研究對象,采用Inmembrane方法和COG功能數據庫對預測的外表面蛋白進行功能注釋和聚類分析。[結果]AD011菌體暴露外表面蛋白數目為193個,包括11個細胞壁蛋白、20個脂蛋白、暴露外表面的膜蛋白140個、22個細胞外蛋白。AD011外表面蛋白的功能分析結果顯示,193個外表面蛋白中,含有較大比例的蛋白沒有功能注釋(78個),115個蛋白具有COG功能注釋。在有功能注釋的蛋白中,所占比例較大的是氨基酸轉運與代謝(16個),無機離子轉運與代謝(14個),細胞壁、細胞膜生物合成(13個),碳水化合物轉運與代謝(10個),防御機制(10個)等有關蛋白。[結論]AD011菌體外表面蛋白與營養物質的吸收和轉運,細胞壁、細胞膜生物合成,防御機制有關。

Abstract[Objective]Species and function of the outer surface proteins of Bifidobacterium animalis AD011 were analyzed.Analysis of their outer surface proteins will help us to correctly understand its physiological function and molecular mechanisms of interaction with their host. [Method]The genome sequence of B. animalis AD011 was used to study the potentially surface exposed proteins ( PSE proteins ) by Inmembrane. Functional annotation and cluster analysis of predicted surface proteins were performed using COG functional database. [Result]There were 193 surface exposed proteins which include 11 cell wall proteins, 20 lipoproteins, 140 membrane proteins and 22 extracellular proteins. Results of functional analysis showed there were 78 proteins which have not COG functional annotation in 193 surface exposed proteins. Most outer surface proteins were involved in amino acid transport and metabolism(16 proteins), inorganic ion transport and metabolism(14 proteins),cell wall/membrane formation(13 proteins), carbohydrate transport and metabolisms(10 proteins), defense mechanisms(10 proteins). [Conclusion]The function of outer surface proteins of AD011 were involved in absorption and transport of nutrient, cell wall/membrane biosynthesis and defense mechanisms.
