摘要[目的]運(yùn)用生物信息學(xué)方法預(yù)測(cè)動(dòng)物雙歧桿菌AD011定位于細(xì)胞膜的表面脂蛋白,為后續(xù)研究該菌與宿主相互作用的分子機(jī)制提供基礎(chǔ)資料。[方法]用SignalP 4.0和LipoP軟件對(duì)動(dòng)物雙歧桿菌AD011全基因組編碼蛋白中表面脂蛋白和信號(hào)肽進(jìn)行預(yù)測(cè),同時(shí)采用COG 功能數(shù)據(jù)庫(kù)對(duì)預(yù)測(cè)的脂蛋白進(jìn)行功能注釋和聚類分析。[結(jié)果]對(duì)動(dòng)物雙歧桿菌AD011基因組1 518個(gè)蛋白的表面脂蛋白預(yù)測(cè),結(jié)果表明,19個(gè)蛋白具有脂蛋白信號(hào)肽。功能分析顯示,該菌株19個(gè)脂蛋白中有3個(gè)蛋白沒(méi)有功能注釋,16個(gè)蛋白有功能注釋,其中10個(gè)蛋白參與氨基酸代謝與轉(zhuǎn)運(yùn)、3個(gè)蛋白參與碳水化合物代謝與轉(zhuǎn)運(yùn),3個(gè)蛋白參與無(wú)機(jī)鹽離子代謝與轉(zhuǎn)運(yùn)。[結(jié)論]動(dòng)物雙歧桿菌AD011表面脂蛋白參與的功能主要與營(yíng)養(yǎng)物質(zhì)的轉(zhuǎn)運(yùn)和代謝有關(guān)。

Abstract[Objective] The surface lipoproteins of Bifidobacterium animalis AD011 were predicted by bioinformatics method, which provided the basis for interaction molecular mechanisms between bacteria and their host.[Method] SignalP 4.0 and LipoP softwares were used to analyze the lipoproteins and characteristic of signal peptides. Function of lipoproteins was also analyzed by COG database.[Result] There were 19 proteins which possessed signal peptide of lipoproteins from 1 518 proteins coded by B. animalis AD011 genome.Function analysis revealed that the strain contains 3 hypothetical proteins without function annonation.16 proteins have function annonation which mainly involved in amino acid metabolism and transport(10 proteins), transport of carbohydrate metabolism(3 proteins), metabolism of inorganic ion transport(3 proteins).[Conclusion] Function of surface lipoproteins of B. animalis AD011 are mainly associated with the transport and metabolism of nutrients.

動(dòng)物雙歧桿菌AD011是一種重要的商業(yè)化益生菌株,被認(rèn)為是“GRAS”食品原料[1],它具有較強(qiáng)的耐酸和耐膽汁能力,優(yōu)良的腸道黏附能力。2009年對(duì)該菌株進(jìn)行了全基因組測(cè)序和功能注釋,該菌株基因組全長(zhǎng)為1.933 Mb,1 614個(gè)基因,1 518個(gè)編碼蛋白。該菌株全基因組序列的完成使該菌株的研究進(jìn)入功能基因組學(xué)時(shí)代[2]。
細(xì)菌定位于細(xì)胞膜表面的脂蛋白在適應(yīng)外界環(huán)境,與宿主細(xì)胞發(fā)生相互作用等方面起到非常重要作用,表面脂蛋白引起一些研究者的關(guān)注,研究發(fā)現(xiàn),位于雙歧桿菌表面的脂蛋白BopA與黏附作用有關(guān)[3-5]。……
