摘要:本研究旨在探討肝臟型脂肪酸結合蛋白(L-FABP)基因遺傳變異與貴州白山羊生長性狀的關聯性,以期得到能夠影響貴州白山羊各群體生長性狀顯著效應的分子遺傳標記。試驗采用DNA池、PCR-SSCP法結合PCR產物直接測序對貴州白山羊L-FABP 基因4個外顯子進行SNPs篩選,并利用SPSS 180統計軟件分析H-FABP基因遺傳變異與生長性狀間的相關性。多態位點檢測表明,貴州白山羊L-FABP基因內含子兩處存在C4557T和A4575G兩個多態基因座;遺傳效應分析表明,L-FABP基因C4557T和A4575G位點多態座位有利于提高貴州白山羊體重、體高、體斜長和管圍,其中基因型CCGG有利于提高貴州白山羊成年母羊的體高和體斜長;TTGG有利于提高貴州白山羊周歲母羊的體重和管圍。研究結果初步推測L-FABP基因可能影響貴州白山羊部分生長性狀的主效基因或與主效基因連鎖,其多態座位可能作為山羊生長性狀選擇的有效遺傳標記。

Abstract:The aim of this study was to explore the correlation between the genetic variation of L-FABP gene and the growth traits of Guizhou white goat in order to obtain molecular genetic markers that could affect the goats growth traits. The SNPs of four different exons of L-FABP genes of Guizhou white goat were screened by DNA pooling and PCR-SSCP combined with PCR product direct sequencing. The correlation between the genetic variation of L-FABP gene and the growth traits was analyzed by SPSS (18.0) software. Examination of the polymorphism loci showed that two SNPs (C4557T and A4575G) were presented in the Intron 2 of L-FABP gene. The effect analysis showed that the polymorphic loci of C4557T and A4575G sites of L-FABP gene can improve body weight, body height, body length and pipe circumference. The genotype CCGG can be in favour of improving body height and body length of adult ewes. The genotype TTGG can be in favour of improving body weight and pipe circumference in aging ewes. The results indicated that L-FABP gene would be a potential major gene or a gene linked to other major genes affecting goat growth traits; its polymorphic loci could be used as effective genetic markers for goat growth traits to guide the molecular breeding of goats.

肝臟型脂肪酸結合蛋白(Live fatty acid binding protein, L-FABP)是脂肪酸結合蛋白家族(Fatty acid binding proteins, FABPs)重要組成部分,又稱Z-蛋白,亞鐵血紅素蛋白,是一種與遺傳相關的脂肪酸連接蛋白家族成員,在哺乳動物中,主要表達場所是肝臟組織和小腸,在人的腎臟中也有少量表達[1]。L-FABP基因由4個外顯子和3個內含子組成,有127個氨基酸殘基和2個脂肪酸結合位點,分子量約為144 kDa[2]。L-FABP與FABPs家族成員間氨基酸序列的同源性達3870%,主要由核苷……
