摘要:采用柱前衍生化HPLC法測定了12批鐵皮石斛破壁飲片中甘露糖的含量。藥材采用80%乙醇回流提取除雜后,水煎煮制得石斛多糖,然后將多糖經酸水解為單糖,再用1-苯基-3-甲基-5-吡唑啉酮( PMP)進行衍生化處理,以鹽酸氨基葡萄糖作為內標進樣測定。結果平均校正因子為08248,RSD為063%,平均加樣回收率為9803%。每批鐵皮石斛破壁飲片中單糖含量存在明顯差異,但均符合藥典要求。

Abstract:Pre-column derivatization HPLC method was adopted to measure mannose content in 12 batches of Dendrobium. After removing impurity by reflux extraction with 80% ethanol, Dendrobium polysaccharides of the medicinal materials were prepared by water decoction, and then hydrolyzed into monosaccharides which was treated with 1- phenyl -3- methyl -5- (PMP). Glucosamine hydrochloride was used as the internal standard for sample determination. The results showed that the average correction factor was 08248, RSD was 063%, and the average recovery was 9803%. Although significant differences in the content of monosaccharide in each batch of medicinal materials were observed, they all met the pharmacopoeia requirements..

鐵皮石斛藥材是蘭科植物鐵皮石斛Dendrobium officinale Kimura et Migo的干燥莖,具有益味生津,滋陰清熱的功效[1],現代藥理學研究表明,鐵皮石斛多糖具有增強免疫力、降血糖、降血壓、抗氧化、抗腫瘤等藥理作用[2-5],為其主要藥效成分。經研究,鐵皮石斛多糖主要由甘露糖、半乳糖醛酸、葡萄糖、半乳糖、木糖和阿拉伯糖組成,其中甘露糖的含量最高[6-7]。破壁飲片是采用現代超微技術,將中藥飲片粉碎至300目、粒徑小于45微米,可供直接口服使用的新型中藥飲片。中藥破壁飲片具有高效免煎、攜帶方便、質量可控、安全衛生等特點,因此成為近年中藥研究的熱點[8]。
本實驗采用柱前衍生化HPLC法,測定了貴州某企業提供的12批鐵皮石斛破壁飲片中甘露糖的含量。為鐵皮石斛破壁飲片的質量控制提供了依據,對石斛多糖的活性研究具有進一步的意義。
