以10 mol%三乙烯二胺為催化劑,3-異硫氰酸酯氧化吲哚與α-羰基磷酸酯經(jīng)aldol/環(huán)化反應(yīng),合成了一系列新型的螺[氧化吲哚-3,4'-噁唑啉]-5'-磷酸酯類化合物,收率86%~92%,dr>99∶1,其結(jié)構(gòu)經(jīng)1H NMR 和13C NMR表征。

螺環(huán)氧化吲哚結(jié)構(gòu)單元廣泛存在于許多天然分子中,已發(fā)現(xiàn)的含3,3'-螺環(huán)結(jié)構(gòu)的氧化吲哚類化合物大多具有較高的生物活性,如抗病毒、抗細(xì)胞毒素、抗氧化等[1-2]。如Spirobrassinin及其類似物Methoxyspirobrassinin(Chart 1)是一類植物抗毒素氧化吲哚,具有殺菌、抗腫瘤活性[3]。而含磷化合物由于其獨(dú)特的醫(yī)藥價(jià)值,受到化學(xué)家們?cè)絹碓蕉嗟年P(guān)注,如Estramustine phosphate(Chart 1)對(duì)前列腺癌具有明顯的治療作用[4]。鑒于含磷化合物與螺環(huán)氧化吲哚類化合物廣泛的生理與藥物價(jià)值,構(gòu)建此類化合物具有重要的意義。

本課題組[5-7]曾報(bào)道了3-異硫氰酸酯氧化吲哚(1)的合成,并研究了1與醛、酮、亞胺及不飽和雙鍵的加成環(huán)化反應(yīng),但是1與α-羰基磷酸酯的反應(yīng)還未見報(bào)道。根據(jù)本課題組多年的研究基礎(chǔ)預(yù)測(cè)1與α-羰基磷酸酯可發(fā)生aldol/環(huán)化反應(yīng),用于構(gòu)建含磷酸酯的螺環(huán)氧化吲哚類化合物。

本文以10 mol%三乙烯二胺(DABCO)為催化劑,1和α-羰基磷酸酯(2a~2h)經(jīng)aldol/環(huán)化一步構(gòu)建含磷酸酯的螺環(huán)氧化吲哚類化合物(3a~3h);由于3不太穩(wěn)定(可能發(fā)生逆aldol反應(yīng)),通過一步簡(jiǎn)單的甲基化反應(yīng),將其轉(zhuǎn)化為另一類穩(wěn)定的新型螺[氧化吲哚-3,4'-噁唑啉]-5'-磷酸酯類化合物(4a~4h,Scheme 1),收率86%~92%,dr>99∶1,其結(jié)構(gòu)經(jīng)1H NMR和13C NMR表征。

1 實(shí)驗(yàn)部分

1.1 儀器與試劑

Bruker-300型核磁共振儀(CDCl3為溶劑,TMS為內(nèi)標(biāo))。

所用試劑均為分析純。

1.2 合成

(1)4的合成通法

在硬質(zhì)反應(yīng)管中加入DABCO 2.2 mg(0.01 mmol),2 0.2 mmol和THF 3 mL,冰浴冷卻下攪拌3 min;加入1 28.0 mg(0.1 mmol),反應(yīng)30 min (TLC監(jiān)測(cè))。減壓旋蒸除去THF后經(jīng)硅膠柱層析[洗脫劑:A=V(乙酸乙酯)∶V(石油醚)= 1∶2~1∶1.5]純化得3。

將3溶于4 mL丙酮中,依次加入K2CO316.5 mg(0.12 mmol)和MeI 17 mg(0.12 mmol),攪拌下于室溫反應(yīng)過夜。減壓蒸除丙酮后經(jīng)硅膠柱層析(洗脫劑:A=1∶3~1∶2)純化得無色油狀液體4。

2 結(jié)果與討論

2.1 合成

在4的合成中,2中的Ar無論是吸電子取代基(2d,2e和2h)還是給電子取代基(2b和2c),對(duì)反應(yīng)活性基本無影響,均可高收率合成4;當(dāng)Ar 的2-位為吸電子取代基(2d和2e)時(shí),反應(yīng)的非對(duì)應(yīng)選擇性(dr)大大降低;對(duì)于雜芳環(huán)和大位阻的萘環(huán)(2g),該反應(yīng)體系也有很好的適應(yīng)性,只是對(duì)于芳雜環(huán),dr有所降低(2f),而Ar為萘環(huán)時(shí)收率89%,dr>99∶1。

2.2 反應(yīng)機(jī)理

推測(cè)合成4的機(jī)理如Scheme 2所示。

3 結(jié)論

以10 mol%三乙烯二胺為催化劑,通過aldol/環(huán)化反應(yīng),高收率的合成了一系列螺[氧化吲哚-3,4'-噁唑啉]-5'-磷酸酯類化合物,非對(duì)應(yīng)選擇性>99∶1。該方法為合成含磷螺環(huán)氧化吲哚類化合物提供了很好的途徑。
