電融合法產生駱駝刺與鷹嘴紫云英屬間體細胞雜種

采用電融合法獲得了駱駝刺和鷹嘴紫云英屬間體細胞雜種。駱駝刺原生質體來自發根農桿菌A4轉化系細胞,并經碘乙酰胺處理;鷹嘴紫云英原生質體從甲硫氨酸抗性系細胞分離。雙親及同源融合產物均不能在無激素的篩選培養基上持續分裂,融合后的雜種細胞由于雙親生理互補效應可恢復持續分裂能力而被篩選出來。本實驗優化了電融合參數,如直流脈沖、交流脈沖和脈沖次數。融合產物經培養獲得的雜種細胞系經形態學、染色體數目檢查、生化及隨機擴增多態性DNA分析,鑒定出10個雜種克隆,并從3個雜種克隆再生了小植株。

Abstract:We obtained intergenus somatic hybrid betweenAlhagi pseudalhagiandAstragalus cicerby using electroporation.Agrobacterium rhizogenesA4-transformedA. pseudalhagiprotoplasts were treated with iodoacetamide so that they were unable to sustain divisions.A. cicerprotoplasts were isolated from a methionine-resistant mutant and did not survive in the medium without plant growth regulator. The intergenus somatic hybrid cells were selected based on physiological complementation between the two parents. We optimized some parameters of electroporation, such as direct current impulse, alternating current impulse and the impulse number. We identified ten hybrid clones by morphological observation, checking the chromosome numbers, isoenzyme analysis and random amplified polymorphic DNA analysis, and obtained regenerated plantlets from three hybrid clones.

通過原生質體融合和培養產生體細胞雜種是克服有性不親和性、實現遠緣遺傳重組、轉移多基因控制性狀的可行途徑。植物體細胞雜交是植物生物技術的重要組成部分。隨著技術手段的不斷改進,在植物種內、種間、屬間甚至科間的原生質體融合都已成功。有一些優良的性狀,特別是野生種的抗病[1]、抗旱[2]和胞質雄性不育[3]等性狀已通過體細胞雜交成功地轉移到栽培植物中。……
