根癌農桿菌介導的增強型綠色熒光蛋白基因在淡紫擬青霉中的轉化

為了實現增強型綠色熒光蛋白基因 (egfp) 在生防真菌淡紫擬青霉 9410菌株中的轉化,借助中間質粒pcDNA3.1(-) 構建nptⅡ-egfp融合基因的表達載體pUPNGT,然后采用根癌農桿菌介導的轉化法將egfp基因轉化到淡紫擬青霉9410菌株中。PCR檢測和Southern blotting分析結果表明,egfp基因以單拷貝形式整合到淡紫擬青霉9410的基因組中。熒光顯微鏡觀察結果顯示,轉化子在488 nm光源的激發下能產生綠色熒光。這些結果說明egfp基因已成功轉化至淡紫擬青霉 9410菌株并獲得表達。這些工作可為淡紫擬青霉在不同條件下的防效評價、環境安全評價等提供新的途徑和方法。

Abstract:The main aim of this study was to transform the enhanced green fluorescent protein gene (egfp) into biocontrol fungusPaecilomyces lilacinusstrain 9410. We constructed the expression vector pUPNGT of the fusion genenptII-egfpusing pcDNA3.1(-)as a helper plasmid. Theegfpgene was then transformed intoP. lilacinusstrain 9410 viaAgrobacterium tumefaciens-mediated transformation. PCR and Southern blotting analysis showed that theegfpgene was integrated into the genomes of the tested transformants and the integration manner was single-copy. The transformants could generate green fluorescence when they were excited by 488 nm blue laser. These results indicated that theegfpgene had been successfully transformed intoP. lilacinus9410 and expressed in the tested transformants. Our work may provide a new approach to assess environmental safety and practical biocontrol efficacy ofP. lilacinusunder different conditions.

淡紫擬青霉Paecilomyces lilacinus(Thom.)Samson 1974是一種土壤及多種植物根際的習居菌,在全球廣泛分布。現已認為它是最有前途的植物病原線蟲的生防真菌之一,全球已有60多個國家開展了應用淡紫擬青霉防治根結線蟲Meloidogynespp.、胞囊線蟲Heteroderaspp.等植物病原線蟲的研究,并已開始付諸實踐[1-3]。同時,淡紫擬青霉對有害昆蟲[4]、動物寄生蟲[5]、植物病原菌[6]等也有一定的防控效果。但在淡紫擬青霉的遺傳背景及其控害的作用機制等方面,至今仍了解甚少。絲狀真菌遺傳轉化系統的建立,為人們從分子水平了解絲狀真菌的遺傳背景和進一步的菌株改造提供了強有力的工具。……
