






摘要 [目的]建立氣相色譜-質(zhì)譜法(GC-MS)同時測定食用油中β-谷甾醇、豆甾醇、菜油甾醇、菜籽甾醇含量的方法。[方法]樣品選用氫氧化鉀-乙醇皂化、正己烷萃取、濃縮、衍生化后以正己烷定容,采用GC-MS選擇離子掃描模式(SIM),外標(biāo)法定量。[結(jié)果]β-谷甾醇、豆甾醇、菜籽甾醇的線性范圍為5~500 μg/mL,菜油甾醇的線性范圍為2.5~250.0 μg/mL,線性相關(guān)系數(shù)(R2)均大于0.999,檢出限為0.05~0.10 mg/kg,定量限為0.1~0.3 mg/kg,加標(biāo)回收率為80.5%~107.8%,相對標(biāo)準(zhǔn)偏差(RSD)為2.3%~9.8%。[結(jié)論]該方法操作簡單、高效、靈敏度高、重復(fù)性好,適用于食用植物油樣品中β-谷甾醇、豆甾醇、菜油甾醇、菜籽甾醇含量的測定。

Determination of Various Phytosterols in Edible Vegetable Oils by Gas Chromatography-mass Spectrometry

Abstract [Objective]A method for simultaneous determination of β-sitosterol,stigmasterol,campesterol and brassicasterol in edible oil was established by gas chromatography-mass (GC-MS).[Method]Sample was saponified by potassium hydroxide-ethanol solution,n-hexane extraction,concentration,derivatization,and then diluted with n-hexane.GC-MS was used to select ion scanning mode (SIM) and external standard method was used for quantification.[Result] There were good linear relationships for β-sitosterol,stigmasterol,brassicasterol in 5-500 μg/mL,and campesterol in 2.5-250.0 μg/mL,respectively.Linear correlation coefficients(R2)were greater than 0.999.Limits of detection were 0.05-0.10 mg/kg and limits of quantification were 0.1-0.3 mg/kg,respectively.Adding standard recovery rate was 80.5%-107.8%,relative standard deviation (RSD) was 2.3%-9.8%.[Conclusion]This method is easy to operate,efficient,highly sensitive,and has good repeatability,which is suitable for the determination of β-sitosterol,stigmasterol,campesterol and brassicasterol contents in edible vegetable oil samples.

植物甾醇,又被稱為植物固醇,在各類植物中普遍存在,現(xiàn)已被廣泛應(yīng)用在食品、藥品領(lǐng)域。植物甾醇在結(jié)構(gòu)、生化特性上均與膽固醇相似,但又略有不同[1-2]。研究發(fā)現(xiàn),植物甾醇能夠降低人體血清膽固醇(TC)與低密度脂蛋白膽固醇(LDL-C)水平,可以有效防治心血管疾病并加速膽固醇的代謝降解。人體并不能自主合成植物甾醇,但在植物源品中植物甾醇是生物活性成分,人體可從日常飲食中獲得所需的植物甾醇。根據(jù)中華人民共和國衛(wèi)生部公告2010年第3號規(guī)定,在除嬰幼兒食品外,其他各類食品均可以使用植物甾醇,且食用量≤2.4 g/d[3];《中國居民膳食營養(yǎng)素參考攝入量》中推薦植物甾醇攝入量在0.9 g/d[4]。
食用植物油作為人們?nèi)粘5氖秤檬称罚渲泻胸S富的植物甾醇、礦物質(zhì)、不飽和脂肪酸等,同時也是植物甾醇含量較為豐富的食品之一。……
