摘 要:本研究采用高效液相色譜法,同時檢測保健食品中VB1、VB2、煙酸、煙酰胺、VB6(吡多胺、吡哆醛、吡哆辛)、泛酸鈣、VB12、VC共10種水溶性維生素。樣品經Agilent C18柱(4.6mm×250mm,5μm)分離,使用PDA檢測器,流動相為甲醇與0.025mol/L磷酸二氫鉀溶液(加磷酸調節pH=3.1),梯度洗脫。結果表明,10種水溶性維生素得到良好分離,分離度均大于1.5。10種水溶性維生素線性范圍為0.2~50μg/mL,相關系數為0.999 5~1.000 0,平均回收率為92.7%~103.7%,[RSD]為0.37%~2.80%([n]=3);10種水溶性維生素的檢測限為1.5~9.0mg/kg。該方法簡單、方便、快速且準確,可同時測定保健食品中10種水溶性維生素。

Simultaneous Determination of 10 Water-soluble Vitamins
in Health Foods by HPLC

Abstract: In this study, high performance liquid chromatography (HPLC) was used to detect the contents of VB1, VB2, nicotinic acid, nicotinamide, VB6 (pyridoxamine, pyridoxal, pyridoxine), calcium pantothenate, VB12 and VC 10 kinds of water-soluble vitamins. The samples were separated on an Agilent C18 column (4.6mm×250mm , 5 μm) using a PDA detector. The mobile phase was eluted with methanol and 0.025mol/L potassium dihydrogen phosphate solution (adjusted to pH=3.1 with phosphoric acid). The results showed that 10 kinds of water-soluble vitamins were well separated, the separation degree was more than 1.5. 10 kinds of water-soluble vitamins linear range of 0.2~50μg/mL, the correlation coefficient of 0.999 5~1.000 0, the recovery rate of 93% to 103%,[RSD] 0.37%~2.80%([n] = 3). The detection limits of 10 kinds of water-soluble vitamins were 1.5~9.0mg/kg. The method is simple, convenient, fast and accurate, and can simultaneously measure 10 kinds of water-soluble vitamins in health food.

維生素是一類必須從食物中攝取的有機化合物,多為酸、醇和酚類物質。維生素的種類很多,按溶解性可分為脂溶性維生素(A、D、E、K)和水溶性維生素(B族維生素和VC)。B族維生素有12種以上,被世界一致公認的有9種。維生素B族包括維生素B1、維生素B2、維生素B3(煙酸、煙酰胺)、維生素B5(泛酸鈣)、維生素B6(吡哆辛、吡哆醛、吡多胺)、維生素B12(鈷胺素)、維生素B9(葉酸)、維生素B7(生物素)等[1]。
隨著復合維生素制劑及高檔營養品的出現,同時檢測食品藥品中水溶性維生素的含量引起人們的廣泛關注。現行標準分析方法多為微生物法、分光光度法,這類方法耗時、費力,已難以滿足當前分析要求和發展趨勢;也有采用HPLC測定水溶性維生素的方法,但大多限于單一維生素的測定;僅有《保健食品的高效液相色譜測定方法》(GB/T 5009.197—2013)同時測定保健食品中VB2、VB6、煙酸、煙酰胺等4種水溶性維生素,但該方法使用離子對試劑、對色譜柱消耗較大,且儀器難以平衡。……
