【摘要】 目的:探討角質(zhì)細(xì)胞生長(zhǎng)因子(KGF)對(duì)人齦上皮細(xì)胞在鈦金屬表面附著的影響效果。方法:取阻生牙拔除術(shù)患者的健康齦組織塊,經(jīng)過(guò)相關(guān)處理獲取齦上皮細(xì)胞,將該細(xì)胞在純鈦片表面接種,在重組KGF濃度調(diào)配下處理,進(jìn)而計(jì)數(shù)純鈦片表面附著的細(xì)胞數(shù),測(cè)定純鈦片表面細(xì)胞的粘附力,比較接種后6 h、1 d、5 d、10 d、15 d時(shí)不同KGF濃度下純鈦片表面附著的齦上皮細(xì)胞數(shù)目,記錄KGF濃度1 ng/mL與5 ng/mL在培養(yǎng)1 d、10 d時(shí)齦上皮細(xì)胞在純鈦片表面粘附力與平均熒光強(qiáng)度(AFI)。結(jié)果:接種后同一時(shí)間點(diǎn),隨著KGF濃度升高,齦上皮細(xì)胞數(shù)目明顯增多(P<0.05),同一KGF濃度下,隨著接種時(shí)間延長(zhǎng),齦上皮細(xì)胞數(shù)目明顯增多(P<0.05);KGF濃度1 ng/mL與5 ng/mL時(shí),培養(yǎng)1 d與10 d時(shí)齦上皮細(xì)胞在純鈦片表面粘附力與AFI比較,差異均有統(tǒng)計(jì)學(xué)意義(P<0.05),培養(yǎng)10 d時(shí)更高。結(jié)論:隨著KGF濃度升高、接種與培養(yǎng)時(shí)間延長(zhǎng),齦上皮細(xì)胞數(shù)目明顯增加,而且附著力也會(huì)隨著KGF濃度升高而升高,在時(shí)間-濃度變化中,AFI也呈現(xiàn)隨之增高趨勢(shì),可見(jiàn)KGF對(duì)人齦上皮細(xì)胞在鈦金屬表面附著影響較大,需加強(qiáng)重視。

【Abstract】 Objective:To investigate the effect of keratinocyte growth factor(KGF) on adhesion of human gingival epithelial cells on titanium surface.Method:The gingival epithelial cells were obtained from the patients with impacted teeth,and the gingival epithelial cells were obtained by related treatment,they were inoculated on the surface of pure titanium sheets,processed under the concentration of recombinant KGF,the number of cells attached to the surface of pure titanium sheets was counted,and the adhesion force of the cells on the surface of pure titanium sheets was measured.The number of gingival epithelial cells adhered to pure titanium slices at different concentrations of KGF at 6 h,1 d,5 d,10 d and 15 d after inoculation were compared,the adhesive force and average fluorescence intensity(AFI) of gingival epithelial cells on pure titanium sheets at l ng/mL and 5 ng/mL of KGF concentration at 1 d and 10 d were recorded.Result:At the same time point after inoculation,the number of gingival epithelial cells increased significantly with the increase of KGF concentration(P<0.05).At the same KGF concentration,the data of gingival epithelial cells increased significantly with the extension of inoculation time(P<0.05).When the concentration of KGF was 1 ng/mL and 5 ng/mL,the adhesion force and AFI of gingival epithelial cells on pure titanium surface were compared at 1 d and 10 d,the differences were statistically significant(P<0.05),and the higher at 10 d.Conclusion:With the increase of KGF concentration,inoculation and incubation time,the number of gingival epithelial cells increased significantly,and the adhesion also increased with the increase of KGF concentration,in the time-concentration change,AFI also showed a trend of increasing,it showed that KGF had a great influence on the attachment of human gingival epithelial cells on the surface of titanium,so we should pay more attention to it.endprint

種植體表面的上皮附著是種植牙成功的關(guān)鍵。阻生齒拔除后口腔表面上皮可能往牙面爬行生長(zhǎng),而且重新分化結(jié)合生皮,或者出現(xiàn)新的結(jié)合上皮,并可分泌出基底膜,形成新的上皮附著,稱之為上皮再附著,組織主要源于口腔齦上皮[1-3]。隨著人們對(duì)齦上皮細(xì)胞的了解增多,臨床對(duì)其興趣也逐漸增加,尤其是其在鈦金屬表面的附著情況[4-6]。角質(zhì)細(xì)胞生長(zhǎng)因子(KGF)被證實(shí)有著促進(jìn)上皮細(xì)胞增生的效果[7-9],為了進(jìn)一步探討KGF對(duì)人齦上皮細(xì)胞在鈦金屬表面附著的影響情況,本文進(jìn)行了如下研究,現(xiàn)報(bào)道如下。
1 材料與方法
1.1 細(xì)胞來(lái)源與鑒定
1.1.1 來(lái)源 選擇阻生齒拔除患者健康齦組織塊,利用4 ℃、0.25%分離酶進(jìn)行16 h作用,分離齦上皮組織及其下結(jié)締組織,采取0.125%胰酶消化上皮片形成一種單一的人齦上皮細(xì)胞單細(xì)胞懸液,進(jìn)行體外原代培養(yǎng)、傳代培養(yǎng),每天利用倒置顯微鏡對(duì)其細(xì)胞形態(tài)實(shí)施觀察與記錄。……
