摘 要 利用固相微萃取(SPME)法萃取香草蘭中揮發和半揮發性的成分,再結合全二維氣相色譜/飛行時間質譜(GC×GC/TOFMS)技術進行定性分析。試驗共鑒定出香草蘭中181種揮發、半揮發性化合物,其中含量較高的成分主要有香蘭素、呋喃甲醛、(2S,3S)-(+)-2,3-丁二醇、鄰甲氧基苯酚、2,3-丁二醇、苯酚、正己醛、(2R,3R)-(-)-2,3-丁二醇、對羥基苯甲醛、3-甲基丁酸、2,3,5,6-四甲基吡嗪、3-羥基-2-丁酮、香草酸、壬醛、正己酸、1-辛烯-3-酮。結果表明,GC×GC/TOFMS高分辨率和高靈敏度的特點非常適合復雜組分體系的定性分析。

Abstract Volatile and semi-volatile compounds in vanilla were extracted by using solid phase micro-extraction and were identified by comprehensive two-dimensional gas chromatography/time of flight mass spectrometer. A total of 181 volatile or semi-volatile compounds in vanilla were identified and compounds with high content were vanillin, 2-furancarboxaldehyde,[S-(R*,R*)]-2,3-butanediol, 2-methoxy-phenol, 2,3-butanediol, phenol, hexanal, [R-(R*,R*)]-2,3-butanediol, 4-hydroxy-benzaldehyde, 3-methyl-butanoic acid, tetramethyl-pyrazine, 3-hydroxy-2-butanone, 4-hydroxy-3-methoxy-benzoic acid, nonanal, hexanoic acid, 1-octen-3-one.The results showed that GC×GC/TOFMS with high resolution and sensitivity is suitable for the identification of complex system.

香草蘭(Vanilla planifolia Andrews)是著名熱帶植物,原產于墨西哥,現已引種至中國、馬達加斯加、科摩羅、印度尼西亞、烏干達、印度等地區。香草蘭因其獨特的香味而聞名,作為增香劑廣泛用于食品、冰激凌、化妝品等行業[1-2],因此其也被認為是最具經濟價值的蘭科植物[3]。目前香草蘭香味成分的研究報道較多。Adedeji等[4]利用直接熱力解析法鑒定出香草蘭中61種成分;Perez-Silva[5]利用溶劑萃取法鑒定出墨西哥地區香草蘭中65種揮發性成分;Toth[6]對波旁、塔西提島、烏干達和印度尼西亞地區的香草蘭中進行鑒定比較;Brunschwig等[7]利用同時蒸餾萃取法萃取塔西提島香草蘭中96種揮發性成分。
前人采用不同的萃取方法對香草蘭中揮發性或半揮發性成分進行萃取,但定性方法均為氣相色譜-質譜聯用儀(GC-MS)。全二維氣相色譜/飛行時間質譜(GC×GC/TOFMS)能克服傳統單柱氣相色譜對多組分樣品分離能力相對較差的問題,具有分辨率高、靈敏度高等特點,在分析復雜體系樣品中的應用越來越廣泛[8-10]。……
