摘 要:以嫩芽、莖尖為外植體,研究巨尾桉DH3229的組織培養(yǎng)及育苗技術(shù)。實(shí)驗(yàn)證明,不同的激素組合對(duì)巨尾桉DH3229外植體的誘導(dǎo)、生根有不同的影響。最適合誘導(dǎo)的增殖培養(yǎng)基為MS+6-BA 0.5mg/L+NAA 0.2mg/L,其誘導(dǎo)出來的植株生長(zhǎng)最快速,分枝最多,植株最高;最適合的生根培養(yǎng)基為MS+NAA 1mg/L+6-BA 0.2mg/L,生根率達(dá)94%,主根粗壯,平均長(zhǎng)3根根須。但是移植到基質(zhì)為黃土的培養(yǎng)杯中育苗時(shí),成活率最高的培養(yǎng)基是MS+NAA 0.5mg/L+6-BA 0.2mg/L,成活率達(dá)93%。

Study on Tissue Culture and Genetic Improvement of Eucalyptus grandis×Eucalyptus urophylla DH3229

Abstract:The tender buds were used as explants to study the tissue culture and genetic improvement of Eucalyptus grandis×Eucalyptus urophylla DH3229 in the experiment.The results were as follws: The optimal subculture medium was MS+6-BA 0.5mg/L+NAA 0.2mg/L,in which the plants grew expeditiously and strongly.The optimal rooting medium for test-tube plants was MS+NAA 1mg/L+6-BA 0.2mg/L,in which the rooting rate reached 94% and per plant could produce 3 roots.But the best optimum medium for plant transplantation was MS+NAA 0.5mg/L+6-BA 0.2mg/L,in which the transplanting survival rate of the plants reached 93%.

桉樹(Eucalyptus)為桃金娘科桉樹屬植物的總稱,共有945個(gè)種和變種,原產(chǎn)地為澳大利亞和印度尼西亞及其附近的島嶼[1]。桉樹是世界三大速生樹種之一,也是經(jīng)濟(jì)林和防護(hù)林的理想樹種,而且具有速生豐產(chǎn)、適應(yīng)性廣、抗性強(qiáng)、繁殖容易等特點(diǎn)而被全球廣泛引種[1-3]。巨尾桉(Eucalyptus grandis×Eucalyptus urophylla)優(yōu)株是以巨桉(E·grandis)為母本、尾葉桉(E·urophylla)為父本的雜交種[4]。巨尾桉具有生長(zhǎng)迅速、樹干圓滿通直、輪伐期短的特點(diǎn),是發(fā)展工業(yè)原料林的優(yōu)良闊葉樹種,其木材用途廣,是造紙、造船和膠合板等的主要原料,木材暢銷[5]。
筆者利用不同生長(zhǎng)調(diào)節(jié)劑對(duì)巨尾桉DH3229外植體組織培養(yǎng)及育苗方面進(jìn)行研究,研究結(jié)果為解決巨尾桉快速繁殖大量?jī)?yōu)質(zhì)種苗具有一定的指導(dǎo)意義,并且為實(shí)現(xiàn)規(guī)模化育苗提供理論基礎(chǔ),對(duì)于建立高效、優(yōu)質(zhì)的桉樹外植體組織培養(yǎng)系統(tǒng)等研究也有重要參考價(jià)值。
1 材料與方法
1.1 材料 在廣西玉林市林業(yè)科學(xué)研究所孤木沖的優(yōu)良巨尾桉DH3229品系中,選擇樹干筆直、樹冠高大、生長(zhǎng)良好的植株作為母本,在根部距離地面約20~30cm處進(jìn)行環(huán)割,約100d后,取長(zhǎng)出的嫩芽嫩莖作為外植體。……
