摘要:以水杉(Metasequoia glyptostroboide)種子為材料,設(shè)置全光照、全黑暗和光暗交替(各12 h/d)3種不同光照和室溫、4 ℃干藏的儲(chǔ)藏條件,研究這些條件下水杉種子的萌發(fā)特性、酶活性及可溶性糖變化。結(jié)果表明,在3種光照處理下,水杉種子最適宜的萌發(fā)條件為全黑暗處理。在水杉種子儲(chǔ)藏過(guò)程中,4 ℃干藏條件下的種子POD活性始終高于室溫干藏;4 ℃干藏條件下的種子SOD同工酶變化幅度相對(duì)于室溫儲(chǔ)藏條件小,而兩種儲(chǔ)藏溫度下種子的可溶性糖變化幅度都較小,所以4 ℃是比較適合水杉種子儲(chǔ)藏的溫度。

Influence of Different Light and Storage Temperature on Germination Rate and
Enzyme Activities of Metasequoia glyptostroboide Seeds

Abstract: Metasequoia(Metasequoia glyptostroboide) seeds were used to research germination performance under three kind of different treatments (full illumination, full darkness and alternation of light and darkness every 12 hours, respectively)and two different storage conditions (room tempreture and 4 ℃,respectively). The influence of different storage conditions on metasequoia seeds’ vigor and enzymatic activities were studied. The results showed that full darkness was the most suitable lighting conditions for metasequoia seeds germination among the three light treatments. And the seed germination rate under 4 ℃ storage conditions was higher than that of the room temperature storage condition. The enzymatic activity of peroxidase in metasequoia seeds at 4 ℃ storage condition was higher than that of the room temperature storage condition. However, the variation range of superoxide dismutase enzymatic activity was smaller under 4 ℃ than that under room temperature. The variation range of soluble sugar in metasequoia seeds were smaller at both two storage cinditions. Therefore, 4 ℃ storage conditions was more appropriate for the storage of metasequoia seeds.

水杉(Metasequoia glyptostroboides Hu et Cheng)屬杉科(Taxodiaceae)水杉屬(Metasequoia)植物,中國(guó)特有孑遺珍貴樹(shù)種,被列為國(guó)家一級(jí)保護(hù)植物,素有“活化石”之稱。水杉在中生代白堊紀(jì)及新生代曾廣泛分布于北半球,第四紀(jì)冰期以后,水杉屬的其他種類全部滅絕,僅存水杉一種。水杉野生種僅分布于湖北利川、四川石柱和湖南龍山,對(duì)古植物、古氣候、古地理和地質(zhì)學(xué)以及裸子植物系統(tǒng)發(fā)育的研究均有重要意義[1]。水杉樹(shù)干通直挺拔,生長(zhǎng)快,葉色翠綠,秋后葉色金黃,是庭院觀賞和亞熱帶地區(qū)平原綠化的優(yōu)良樹(shù)種,也是速生用材樹(shù)種[2,3]。
目前,對(duì)水杉的研究主要集中在其地理分布、系統(tǒng)演化地位、群落生態(tài)學(xué)以及遺傳多樣性等方面[4-6]。由于水杉種子天然發(fā)芽率低,種子向幼苗的轉(zhuǎn)型率低,自然更新困難,加上人為破壞等原因,致使野生水杉數(shù)量急劇下降而處于瀕危狀態(tài)[7,8]。……
