摘要:利用蠟狀芽孢桿菌(Bacillus cereus)液體發酵制備γ-聚谷氨酸(Poly-γ-glutamic acid,γ-PGA),提取并制得γ-PGA溶液;以0.5 g/L的γ-PGA溶液與明膠交聯制成明膠膜,未交聯γ-PGA的明膠膜作空白對照,用原子吸收分光光度法檢測金屬離子消耗,研究γ-PGA明膠膜對Cu2+、Co2+的吸附特性。實驗結果表明,Cu2+在pH為8、24 ℃、5 h時有最佳吸附量,飽和吸附量為12.34 μg/mL;Co2+在pH 7、21 ℃、5 h時有最佳吸附量,飽和吸附量為19.50 μg/mL;證實了γ-PGA與明膠交聯制成的明膠膜具有吸附重金屬(Cu2+、Co2+)的能力。

Study on the Adsorption Characteristic of Metal Ions on Gelatin Membrane
Cross-linked Poly-γ-glutamic Acid

Abstract: Poly-γ-glutamic acid (γ-PGA) was produced through liquid fermentation of Bacillus cereus. The cross-linked membrane was prepared by crosslinking γ-PGA(0.5 g/L) with gelatin. The non-cross-linked gelatin membrane was served as control. The adsorption characteristics of Cu2+ and Co2+ on cross-linked membrane were investigated, and the removal quantities of metal ions were measured by atomic absorption spectrophotometry. The results showed that the largest adsorption capacities of Cu2+ was obtained under pH 8 at 24 ℃ for 5 h, and the largest adsorption capacities of Co2+ was obtained under pH 7 at 21 ℃ for 5 h. It was proved that the cross-linked membrane produced with crosslinked γ-PGA and gelatin was capable for the adsorption of metal ions (Cu2+ and Co2+). This study established a research foundation for the recycle of the valuable metals.

γ-聚谷氨酸(Poly γ-glutamic acid)簡稱γ-PGA,γ-PGA是一種由微生物大量生物合成的陰離子型多聚氨基酸,是一種谷氨酸的高分子聚合物,對人類和環境無毒無害[1]。由于其分子量特別大并且側鏈上具有游離的羧基,可以與水中的膠體顆粒相吸附,在適宜的條件下,顆粒之間能夠通過產生“架橋”現象形成一種三維的網狀結構而沉降,可以作為一種新型的生物絮凝劑應用于水的純化和加工以及食品和發酵工業的下游處理[2]。同時,γ-PGA是一種出色的綠色塑料,它在自然界可迅速降解,不會造成環境污染。因而,篩選合適的能產具有絮凝活性的γ-PGA產生菌是當前的研究熱點之一。迄今已經發現了幾種產生菌,主要是芽孢桿菌屬(Bacillus)的菌株,包括各種枯草芽孢桿菌(Bacillus subtilis)[3]、地衣芽孢桿菌(Bacillus licheniformis)[4]、炭疽芽孢桿菌(Bacillus anthracis)[5]和納豆芽孢桿菌(Bacillus natto),但對于蠟狀芽孢桿菌(Bacillus cereus)發酵產生γ-PGA及其絮凝特性的研究鮮見報道。
本實驗通過自行篩選獲得了一株產γ-PGA的蠟狀芽孢桿菌HYWL-1,對其所產的γ-PGA進行了對重金屬離子吸附特性的研究。……
