摘要:為了優(yōu)化101澄清劑對三七(Panax notoginseng)莖葉水提取液的澄清方法,在單因素分析的基礎(chǔ)上應(yīng)用正交設(shè)計考察了101澄清劑組分A、B的加入順序、用量、溫度、作用時間對澄清劑作用效果的影響。得出三七莖葉水提液的最佳澄清條件為先加入0.6 g/L 101澄清劑組分A,再加入1.2 g/L組分B;每加入一種組分后在60 ℃下作用20 min即可達到澄清除雜的目的。

Optimization of Clarifying Conditions of 101 Natural Clarificant for Water Extract from Stems and Leaves of Panax notoginseng

Abstract: 101 natural clarificant was used to clarify water extract from the stems and leaves of Panax notoginseng(Burk.) F.H.Chen. The effects of adding order of 101 clarifiers A and B, amount of clarifiers, clarifying temperature and time on the clarification were studied by single factor test and orthogonal design. The optimum clarifying conditions of water extract from the stems and leaves of P. notoginseng by 101 clarificant were obtained as follows, adding 0.6 g/L clarifier A then adding 1.2 g/L clarifier B, reacting for 20 minutes at 60 ℃ after adding each clarifier.

中藥水提取液成分復(fù)雜,既有有效成分,也有無效成分。有效成分的分子量較小,一般在1 000以下,無效成分的分子量則較大,一般在5 000以上。如果不設(shè)法去除這些高分子雜質(zhì),往往會影響提取分離效果。三七(Panax notoginseng)莖葉中含有皂苷、黃酮類等多種活性成分,但是淀粉、蛋白質(zhì)、多糖等雜質(zhì)常常會對活性成分的提取造成干擾。傳統(tǒng)的水提醇沉法雖然可以很好地沉淀雜質(zhì),但是其乙醇用量較大,對中藥提取液要求濃縮到一定程度,試驗成本高、操作復(fù)雜,且有乙醇殘留[1,2]。101澄清劑是一種新型食用果汁澄清劑,主要成分為變性淀粉[3],安全、無味、無毒,不會引入任何雜質(zhì),并可隨沉淀后的不溶物質(zhì)一并濾去[4]。其操作簡單,無殘留,澄清效果明顯,可以用來澄清三七莖葉的水提取液,本試驗對三七莖葉水提液的101澄清工藝進行了優(yōu)化,以為其工藝改進提供一定的參考。
1材料與方法
1.1材料與儀器
三七莖葉由云南文山三七研究所提供。101澄清劑,購自泰州恒泰器化玻有限公司,組分A用蒸餾水配制成質(zhì)量分數(shù)為1%的溶液并溶脹24 h,組分B用1%醋酸溶液配制成質(zhì)量分數(shù)為1%的溶液并溶脹24 h后用雙層紗布過濾。……
