摘要:構建了合浦珠母貝(Pinctada fucata)的cDNA文庫,并通過EST序列篩選和重新測序獲得了一個新的C型凝集素基因的cDNA序列,命名為PoLEC2,其長度為1 659 bp,5’端非編碼區(qū)為39 bp,3’端非編碼區(qū)為45 bp,開放閱讀框為1 575 bp,可編碼524個氨基酸;PoLEC2蛋白的分子量約58.0 kD,等電點為5.2;PoLEC2蛋白包含一個信號肽序列(M1-A16)、一個MAM-2結構域和一個C型凝集素的糖識別結構域。同源性分析結果表明,PoLEC2與其他物種C型凝集素的糖識別結構域序列的同源性在26.5%~33.6%,相似性在45.1%~55.6%。組織表達分析表明,PoLEC2基因在肝胰臟、鰓、外套膜、性腺及腸組織中均有表達,經(jīng)溶藻弧菌刺激后8h在腸組織中的表達量顯著上調。

Sequence and Expression Analysis of a Novel C-type Lectin Gene from Pearl Oyster Pinctada fucata

Abstract: The cDNA library was constructed from the pearl oyster(Pinctada fucata). A C-type lectin gene, named as PoLEC2, was obtained by screening and sequencing using EST sequences. The length of PoLEC2 cDNA sequence was 1 659 bp. Sequence analysis showed that the open reading frame of PoLEC2 gene was 1575 bp. The 5’-UTR length was 39 bp and 3’-UTR length was 45 bp. The PoLEC2 gene encoded a protein containing 524 amino acids, and the predicted molecular weight of PoLEC2 protein was about 58.0 kD with the isoelectric point of 5.2. The protein sequence of PoLEC2 gene contained a signal peptide sequence with 16 amino acids (M1-A16), a MAM-2 domain and a carbohydrate recognition domain of C-type lectin. Sequence analysis showed that the identity and similarity of the carbohydrate recognition domain of PoLEC2 genes between the P. fucata and other organisms were 26.5%~33.6% and 45.1%~55.6% respectively. Tissue expression analysis showed that the PoLEC2 gene was expressed in all tissues of P. fucata, and there was maximum expression in the intestine after 8 hours of the bacterial infection.

C型凝集素為一類鈣離子依賴活性的糖蛋白,具有至少一個糖識別結構域(Carbohydrate recognition domains,CRD),能夠識別和結合細胞膜表面的糖基,其結合糖基具有專一性,即一種凝集素只能結合一種糖基[1,2]。C型凝集素是一種模式識別受體(Pattern recognition receptor,PRR)蛋白,主要有Man型配體(Man-type ligands)和Gal型配體(Gal-type ligands),Man型配體與D-甘露糖、D-葡萄糖結合,Gal型配體與D-半乳糖及其衍生物結合[2]。PRR蛋白能識別病原微生物引起宿主的免疫應答,在無脊椎動物免疫防御中發(fā)揮著重要作用[1,2]。
合浦珠母貝(Pinctada fucata)又稱馬氏珠母貝,是我國海水珍珠的主要種類,由其所產(chǎn)的珍珠即為舉世聞名的“南珠”。近年來,由于養(yǎng)殖技術落后、養(yǎng)殖環(huán)境污染和種質退化,致使養(yǎng)殖合浦珠母貝出現(xiàn)了生長緩慢、病害頻發(fā)及死亡率高等現(xiàn)象[3],嚴重影響了合浦珠母貝養(yǎng)殖業(yè)的發(fā)展。因此,必須加強合浦珠母貝免疫防御分子機制的研究,為其免疫防治策略的制定提供理論依據(jù)。……
