







摘要:從采集的造紙廠土樣中,篩選出l株產(chǎn)堿性纖維素酶酶活力較高的菌株H-1,經(jīng)鑒定,該菌株為革蘭氏陽(yáng)性芽孢桿菌屬。發(fā)酵產(chǎn)酶條件和酶學(xué)性質(zhì)研究表明H-1菌株的適宜發(fā)酵條件為:接種量為6%,pH 9.0,溫度為37 ℃,此條件下的酶活力可達(dá)8.69 U/mL,是初始酶活力(2.93 U/mL)的3倍。該酶最適反應(yīng)溫度為40 ℃,最適反應(yīng)pH 9.0。在25~55 ℃,pH 7.0~11.0仍能保持60%以上的酶活力,能較好地滿足日常洗滌的環(huán)境要求。

Screening of Alkaline-Cellulose Producing Strain and Study on Its Enzymaic Characterization

Abstract: From soil samples collected from paper mills, one strain called H-1 capable of degrading cellulose was screened and identified as Gram-positive Bacillus. The optimum enzyme-producing conditions were as follows, inoculum amount 6%, pH 9.0 , temperature 37 ℃. The enzyme activity was up to 8.69 U/mL, which was 3 times higher than the initial enzyme activity(2.93 U/mL). The optimal conditions for the enzymatic reaction were about 40 ℃ and pH 9.0. The enzyme activity could be maintained above 60% at temperature 25~55℃ and pH 7.0~11.0, which meet the environmental requirements of the daily washing.

纖維素酶的研究一般都以有效利用纖維素資源為主,酶作用的pH多為酸性,當(dāng)添加到洗滌劑中,由于所處環(huán)境為堿性而失去活力,不能發(fā)揮作用[1]。近年來堿性纖維素酶的發(fā)現(xiàn)使其在洗滌工業(yè)中得到成功應(yīng)用。堿性纖維素酶是纖維素酶系的一個(gè)組分,即羧甲基纖維素酶(CMC酶),它不含有纖維素酶系的其他兩個(gè)組分(外切-β-1,4-葡聚糖苷酶和β-葡萄糖苷酶),能作用于底物CMC(一般用羧甲基纖維素的鈉鹽)。CMC屬大分子多糖衍生物,它被降解后生成多種相對(duì)分子質(zhì)量不等的低聚糖。堿性纖維素酶在對(duì)天然纖維素(棉纖維)作用的過程中不能發(fā)生酶組分間的協(xié)同效應(yīng),不會(huì)使棉纖維受到較多的分解破壞[2,3]。它主要對(duì)棉纖維中占10%左右非結(jié)晶區(qū)的纖維素分子起作用,對(duì)結(jié)晶區(qū)纖維素水解活力很低,因此不會(huì)明顯降低棉纖維的牢固度,并能去除侵入衣物內(nèi)部的塵泥皮脂,顯著提高洗滌效果,節(jié)約洗滌活性物用量,減少環(huán)境污染,是目前世界洗滌工業(yè)發(fā)展的趨勢(shì)[4]。……
