







摘要:利用水浸提法提取茶樹菇(Agrocybe cylindracea)多糖,通過單因素試驗考察料水質量比、浸提時間、pH、醇析時乙醇用量等4個因素對茶樹菇粗多糖得率的影響。結果表明,茶樹菇粗多糖的最佳提取條件為料水質量比1∶10、浸提時間2 h、pH 7.5、浸提濃縮液4倍體積的95%乙醇醇析。用乙醚和Sevage法純化茶樹菇多糖,并對其抗氧化性能進行了分析,發現茶樹菇多糖具有良好的抗氧化性能。在試驗濃度范圍內,其總還原能力和對羥自由基、超氧陰離子自由基、脂過氧自由基的清除能力隨多糖濃度的升高而增強,且其在酸性條件下對亞硝酸根離子具有良好的清除能力。

Study on Extraction of Polysaccharides from Agrocybe cylindracea and Its
Antioxidant Activity

Abstract: Polysaccharide was extracted from Agrocybe cylindracea using water as solvent. According to single factor tests, conditions for obtaining high polysaccharide yield were, mass ratio of material to water, 1∶10; Extraction time, 2 h; pH, 7.5 and volume ratio of 95% ethanol to concentrated extract solution for alcohol precipitation, 4∶1. The extracted crude polysaccharide was refined by ether and Sevage reagent and its antioxidant activity was determined. The results showed that polysaccharide of A. cylindracea was with great antioxidant activity, ability of clearing hydroxyl radical, superoxide anion radical, and lipid peroxy radical nitrite ion increased with the increase of polysaccharide dosage in the scope of test concentration; And it also had excellent clearing ability to nitrite ion in acidity condition.

食用菌是一類具有較高營養價值和藥用價值的食物資源[1],近年來國內外已對多種食用菌,如香菇、靈芝、金針菇、燈芯菇、黑木耳等,進行了水溶性多糖的提取純化、分離鑒定及功能研究[3-7]。茶樹菇(Agrocybe cylindracea)隸屬于糞銹傘科田蘑屬[2],目前對茶樹菇多糖提取方法的研究僅見于王紅梅等[8]的熱水浸提法和毛慧玲等[9]的組合浸提法,而對茶樹菇多糖功能的研究更少[10]。
本研究采用單因素試驗對茶樹菇多糖提取工藝中的料水質量比、浸提時間、pH、醇析過程中乙醇用量4個因素進行優化,并分析了純化后多糖的抗氧化性能,旨在為茶樹菇的綜合開發利用提供參考。
