

摘要:從威海近海采得的海洋沉積物中經(jīng)過富集培養(yǎng)和分離篩選,得到一株紫色菌落的海洋光合細菌,編號為D25。利用梅里埃公司細菌鑒定系統(tǒng)對菌株的生理生化特征進行了檢測,API 20E檢測結(jié)果共有ADH、CIT等4項檢測項目呈陽性,ONPG、LDC等16項檢測項目呈陰性。經(jīng)PCR反應(yīng)得到16S rRNA基因序列,16S rRNA基因序列測定和分析結(jié)果顯示該菌株為嗜硫小紅卵菌 (Rhodovulum sulfidophilum)。

Isolation and Identification of Photosynthetic Bacterium D25

Abstract:A purple strain of marine photosynthetic bacteria numbered D25 was isolated from the marine sediments of WeiHai offshore through enrichment culture, isolation and screen. The physiological and biochemical characteristics of strain D25 were tested by New ATB, the results of API 20E test show that 4 items present positive such as ADH and CIT ,16 items present negative like ONPG and LDC. The 16S rRNA gene sequence were cloned by PCR technology,the results of testing and analyzing of 16S rRNA gene sequence identified the strain as Rhodovulum sulfidophilum.

光合細菌(Photosynthetic bacteria,PSB),是一類在自然界中廣泛存在的,能在厭氧條件下進行不放氧的光合作用并且能夠利用光能自養(yǎng)生長,具有復(fù)雜代謝功能的細菌的總稱。其具有固碳、固氮、脫氮、氧化硫化物等功能,是生命演化中的過渡類型,可作為光合作用機理和產(chǎn)氫固氮機理的研究材料[1-3]。由于光合細菌可以廣泛利用多種有機物,具有在不同的條件下進行多種方式的生長代謝的特點,使其在有機廢水處理中得以應(yīng)用,和傳統(tǒng)活性污泥法相比,利用光合細菌處理污水具有動力消耗低、脫氮除磷效果好、設(shè)備規(guī)模小、占地面積小、管理簡易等明顯優(yōu)勢[1]。此外,由于光合細菌具有凈化水質(zhì)和保護水環(huán)境的作用,加之其本身是活蛋白,營養(yǎng)豐富,在水產(chǎn)養(yǎng)殖方面可以促進生長,提高成活率和養(yǎng)殖效果,含有的抗病毒因子及多種免疫促進因子可活化機體的免疫系統(tǒng),因此其在水產(chǎn)養(yǎng)殖方面也得到了廣泛的應(yīng)用[4]。對海洋沉積物中的一株光合細菌D25進行分離并對其生理生化特性進行研究以及16SrRNA 基因序列的測定和分析,經(jīng)鑒定該菌株為Rhodovulum sp.的Rhodovulum sulfidophilum?!?br>
