摘要:以裙帶菜配子體(Undaria pinnatifida)為試驗材料,研究了高溫脅迫對其葉綠素a含量、POD活性、脯氨酸含量、丙二醛含量的影響。結果表明:與20 ℃對照組相比,裙帶菜配子體在高溫脅迫下,試驗組隨著時間的增加葉綠素a含量明顯下降,且30 ℃下葉綠素a含量的降幅明顯大于25 ℃;丙二醛的含量隨著時間的增加呈現逐漸上升的趨勢,且30 ℃下丙二醛含量的增幅明顯大于25 ℃;POD活性在24 h時達到最低,然后略有增加;脯氨酸的含量在高溫下隨著時間的延長呈現逐漸上升的趨勢。該研究結果為進一步揭示裙帶菜抗高溫分子機制和培育抗高溫裙帶菜新品種提供了一定參考。

Effects of High Temperature Stress on Growth and Physiology of Gametophytes of Undaria pinnatifida

Abstract:Some physiological index including the contents of chlorophyll a, proline, MDA and POD activity of gametophytes of Undaria pinnatifida under different temperature were measured. The results showed that all of the measured contents and activity of gametophytes of Undaria pinnatifida had little change under 20 ℃. When the gametophytes were cultured 48 hours at 25℃ or 30℃, the content of chlorophyll a descended. The contents of praline, MDA increased. POD activity declined in initial stage and then increased and was lower than the control. The results provided some references to the mechanism study of Undaria pinnatifida to high temperature resistance and resistant cultivar breeding.

裙帶菜(Undaria pinnatifida)是中國的三大經濟海藻之一。目前,裙帶菜養殖育苗技術相當成熟,通過配子體培養育苗也有很多報道。溫度是裙帶菜配子體生長發育的重要限制環境因素之一。近年來,溫室效應導致全球氣溫上升,植物生產正面臨著高溫逆境的嚴峻挑戰,因此,研究植物在高溫脅迫下的生理生化反應,將有助于在實際生產中減輕高溫對植物的傷害,同時對抗熱脅迫的基礎理論研究也具有重大意義[1]。
關于溫度對裙帶菜配子體影響的報道集中在對其生長發育的影響[2]。李宏基等[3]指出裙帶菜配子體的最適生長溫度為20 ℃。然而高溫對裙帶菜配子體的生理生化影響研究較少,其抵御高溫的機制還不清楚。本研究以裙帶菜配子體為實驗材料,研究在不同培養溫度下其葉綠素a、脯氨酸、丙二醛和POD活性隨時間而變化的情況?!?br>
