




摘 要: 為了探究黃瓜花葉病毒(Cucumber mosaic virus, CMV)復制酶2a蛋白細胞核定位的生物學意義,以本氏煙UNE12蛋白(NbUNE12)為研究對象,通過雙分子熒光互補實驗及病毒誘導的基因沉默方法分析該蛋白與2a蛋白的相互作用情況及其在CMV侵染過程中的作用。結果表明:CMV 2a蛋白與NbUNE12蛋白的相互作用主要集中于細胞核中;下調表達NbUNE12增加CMV的2b缺陷型突變體(CMVΔ2b-GFP)在植物系統葉的病毒含量,表明NbUNE12蛋白能夠與CMV 2a蛋白發生相互作用,并且負調控CMVΔ2b-GFP侵染本氏煙。該研究為CMV 2a蛋白在植物細胞核定位的生物學功能進一步研究奠定理論基礎。

Abstract:To investigate the biological significance of the nuclear localization of the cucumber mosaic virus (CMV) 2a protein, the article, focusing on Nicotiana benthamiana UNE12 protein (NbUNE12), investigated the interaction between 2a and NbUNE12, as well as the functional role of Nb-UNE12 during CMV infection by conducting bimolecular fluorescence complementation experiments and employing the virus-induced gene silencing method. The findings indicate that the interaction between CMV 2a and NbUNE12 predominantly occured within the nucleus. Silencing NbUNE12 enhanced the accumulation of a 2b-deficient CMV mutant (CMVΔ2b-GFP) in the upper systemic leaves, suggesting that NbUNE12 interacted with CMV 2a and acted as a negative regulator of CMVΔ2b-GFP accumulation. This study lays a theoretical foundation for further study of the biological function of CMV2a protein localization in plant nucleus.

0 引 言
黃瓜花葉病毒(Cucumber mosaic virus, CMV)為雀麥花葉病毒科(Bromoviridae)黃瓜花葉病毒屬(Cucumovirus)的重要成員,能侵染寄主植物多達1000種,且分布范圍廣泛,對農業生產造成嚴重危害[1]。CMV的基因組由3條正義單鏈RNA分子組成,分別命名為RNA1-3,其中RNA1和RNA2分別編碼病毒基因組復制相關的1a蛋白和2a蛋白[2]。此外,由RNA2亞基因組RNA4A編碼的2b蛋白是CMV的主要致病因子,參與病毒長距離移動、影響植物抗性等途徑[3]。2b蛋白能克服宿主水楊酸(Salicylic acid,SA)介導的免疫反應,從而抑制SA對病毒移動的影響。SA預處理能夠導致缺失2b蛋白的CMV突變體(CMVΔ2b)失去系統性侵染的能力[4]。
寄主因子與CMV復制相關蛋白相互作用從而影響病毒的復制。普通煙(Nicotiana tabacum)液泡膜上的鋅指蛋白Tsi1相互作用蛋白1(Tsi1-interacting protein 1,Tsip1)與CMV的1a蛋白和2a蛋白均存在相互作用,并且正調控植物對CMV的抗性[5]。……
