




摘要: 植物生長發育由多種激素共同調控,其中生長素是重要的調控激素之一。生長素響應因子(Auxin response factor,ARF)是生長素信號傳導過程中的關鍵因子,它能夠特異性地與生長素應答元件結合,進而影響植物體內生長素響應基因的表達。本研究通過生物信息學方法鑒定到17個油桐ARF基因家族成員,分布在9條染色體上,并且各基因間無片段重復現象。理化性質分析結果表明,ARF蛋白長度為587~1 119 aa,均為酸性蛋白質,不穩定系數>40,為不穩定蛋白質,親水性指數<0,為親水性蛋白質。根據系統進化分析結果將油桐ARF蛋白家族分為4個亞族,與擬南芥的亞族劃分一致。每個成員都具有典型的B3型結構域和Auxin_resp中間結構域。分析ARF基因表達特征發現,ARF基因的表達在油桐不同組織中具有明顯的組織特異性。本研究結果為揭示油桐生長發育調控機制提供了理論依據。

Identification and expression analysis of ARF gene family in Vernicia fordii

Abstract: Plant growth and development is regulated by many hormones, among which auxin is one of the important regulatory hormones. Auxin response factor (ARF) is a key factor in the process of auxin signal transduction, which can specifically bind to auxin response elements and affect the expression of auxin response genes in plants. In this study, 17 members of ARF gene family in Vernicia fordii were identified by bioinformatics method, which were distributed on nine chromosomes, and there was no fragment duplication between genes. The results of physical and chemical properties analysis showed that the length of ARF proteins was 587-1 119 aa, all of them were acid proteins. The instability coefficients of ARF proteins were more than 40, and the ARF proteins were unstable proteins. The hydropathicity indexes of ARF proteins were less than 0, and the ARF proteins were hydrophilic proteins. According to the results of phylogenetic analysis, the ARF protein family of Vernicia fordii was divided into four subfamilies, which was consistent with the subfamily division of Arabidopsis thaliana. Each member had a typical B3 domain and an auxin_resp intermediate domain. By analyzing the expression characteristics of ARF gene, it was found that the expression of ARF gene had obvious tissue specificity in different tissues of Vernicia fordii. The results of this study provide a theoretical basis for revealing the regulatory mechanism of Vernicia fordii growth and development.

油桐(Vernicia fordii)為大戟科(Euphorbiaceae)油桐屬落葉喬木樹種,其整株都具有極高的應用價值,根、葉、花、果均可入藥[1];并且油桐既有能源和經濟價值,又是重要的用材樹種[2]。
植物生長素響應因子(Auxin response factor,ARF)是生長素信號傳導過程中的關鍵因子,其能夠特異性地結合生長素應答元件,從而影響植物體內生長素響應基因的表達[3]。植物生長素調控涉及多個基因家族,機制十分復雜,對生長素響應因子的研究有助于揭示植物生長發育調控機制。ARF基因在多種植物中的功能已被揭示。……
