重樓醇提物對三種神經(jīng)膠質(zhì)瘤細胞株的細胞毒性研究

【摘 要】 目的：通過重樓醇提物對三種人神經(jīng)膠質(zhì)瘤細胞株（U251、U87、T98G）進行細胞毒性檢測，觀察重樓醇提物的抗腫瘤活性。方法：乙醇冷浸法對重樓飲片進行提取，采用聚酰胺柱層析分離劃段獲得重樓醇提物的不同部位，通過MTT法對所得部位進行細胞毒性檢測。 結(jié)果：重樓醇提物中有3個分離組分作用于人神經(jīng)膠質(zhì)瘤細胞IC50值小于10μg/mL，對神經(jīng)膠質(zhì)瘤細胞株表現(xiàn)出明顯的細胞毒性作用。 結(jié)論：重樓醇提物中存在抑制神經(jīng)膠質(zhì)瘤細胞株增殖的物質(zhì)，值得進一步開展相關(guān)研究。

Abstract：Objective To explore the cytotoxicity induced by paridis ethanol extract on three kinds of glioma cells （U251，U87，T98G）. Methods The ethanol cold-maceration method is used to extract the anti-tumor active components from paridis. The samples are obtained with polyamide column chromatography separation from the paridis ethanol extracts. The cytotoxicity of samples is assessed by MTT assay.Results There are three samples showing strong cytotoxic effect and some of them have the 50% inhibition concentration （IC50） less than 10.0μg/mL. The cytotoxicity analysis shows the paridis ethanol extract having anti-tumor activities. Conclusion Paridis ethanol extract displays an inhibitory effect on these three kinds of glioma cells in vitro and it is worthy further research.

重樓來源于百合科植物云南重樓Paris polyphylla [JP]Smith var.yunnanensis （Franch.）Hand.-Mazz.或七葉一枝花Paris polyphylla Smith var.chinensis （Franch.） Hara的干燥根莖[1]。其性苦、微寒，具有清熱解毒、消腫止痛、涼肝定驚、止咳平喘的功效，在癰腫疔瘡、咽喉腫痛、毒蛇咬傷、抽搐驚風(fēng)、跌打扭傷等癥治療中均有很好的療效。扶正、祛邪是傳統(tǒng)中醫(yī)藥在腫瘤治療方面的兩大基本原則，其中清熱解毒為最主要的祛邪手段之一[2]。重樓組方對惡性腫瘤的熱癥及實癥有一定的治療效果，在惡性腫瘤治療方面的作用越來越受到研究者的關(guān)注，重樓對正常細胞毒性較小、抗腫瘤譜廣，重樓水提物、醇提物及其主要活性成分重樓皂苷和一些重樓單體（如皂苷I、Ⅱ、IV、gracillin、methylmotogracillin、重樓皂苷D等）在體外對食管癌、胃癌、直腸癌、肝癌、胰腺癌、宮頸癌、乳腺癌等多種實體癌均有較強的細胞毒性作用[3]。研究應(yīng)用乙醇冷浸法對重樓飲……
