摘要:為探究蟲生真菌蟬花不同菌株之間產(chǎn)絮凝劑的差異及絮凝劑的絮凝特性,以高嶺土法對36株蟬花菌株進(jìn)行了高產(chǎn)絮凝劑的篩選及高產(chǎn)菌株的生長特性和絮凝性質(zhì)的研究。實(shí)驗(yàn)篩選出穩(wěn)定、高產(chǎn)絮凝劑的蟬花菌株GZUIFR-6722,絮凝劑活性物質(zhì)是其胞外聚合物。該菌株在生長過程中生物量的積累和產(chǎn)絮凝劑能力呈正相關(guān)。當(dāng)菌株6722絮凝劑投加量為12mL每50mL時(shí),絮凝效果較好,絮凝率可達(dá)9147%。1%的CaCl2能顯著促進(jìn)絮凝效果,絮凝率可達(dá)9489%。顛倒次數(shù)、靜置時(shí)間、溫度以及pH對絮凝效果影響不顯著。

Abstract:The aim of this paper is to study the bioflocculability and characteristics of different strains of Isaria cicadae by screening all 36 strains with kaolinThe results showed that GZUIFR-6722 strain can perform steadily with high bioflocculabilityThe active substrate was identified as some kind of extracellular metaboliteDuring the growth,the production of active substrate was related positively with the biomassThe flocculating rate was 9147% after addition of 12mL/50mL bioflocculantsMoreover,the addition of 1% CaCl2 could promote the performance of the bioflocculants,and increased the flocculating rate to 9489%Furthermore,the results also indicated that inversion,standing time,temperature and pH had no obvious effect on flocculationThe findings suggest for application of the GZUIFR-6722 strain in the treatment of waste water

蟬花(蟬棒束孢Isaria cicadae)是一種蟲草類真菌,對蚜蟲、小菜蛾、白粉虱等害蟲的防治效果較好[1,2],同時(shí)也是一味傳統(tǒng)名貴中藥,在糖尿病、抗腎纖維化進(jìn)程、延緩腎衰竭等的臨床及實(shí)驗(yàn)研究中,取得了較好效果[3,4]。迄今為止,對蟬棒束孢透明質(zhì)酸、多糖及糖蛋白等活性物質(zhì)[5-7]和抗衰老、改善免疫功能等[8,9]在醫(yī)療和保健上已有大量的研究報(bào)道,因此,蟬花是一種極具應(yīng)用前景的高安全性的真菌資源。除此之外,也有研究發(fā)現(xiàn)蟬花能產(chǎn)生絮凝劑,郭旭輝等[10]率先發(fā)現(xiàn)蟬花發(fā)酵液具有絮凝活性,采用pH為70的馬鈴薯液體培養(yǎng)基150r/min、30℃搖床培養(yǎng)84h后絮凝率可達(dá)851%。孫嘉龍等[11]從土壤中分離到一株具高效絮凝活性的菌株3716,并對其培養(yǎng)條件與絮凝活性進(jìn)行了研究,由此可見,蟬花的絮凝能力也可用于微生物絮凝劑的開發(fā)。
