摘要以紫色白邊單瓣花的非洲紫羅蘭葉片為試材進行了組織培養技術的研究。結果表明:以葉片為外植體進行組織培養時,愈傷組織誘導最適培養基為MS+0.2 mg/L NAA+0.5 mg/L 6-BA,培養3周左右誘導率達100%;繼代增殖時最適培養基為MS+0.02 mg/L NAA+0.1 mg/L 6-BA;無需生根培養,直接瓶外煉苗,栽植于以m(草炭土)∶m(珍珠巖)=1∶1的基質中生長最好,成活率可達95%以上。

We studied the tissue culture technology ofSaintpauliaIonanthawith the purple petals of white edge and single flower. The tissue culture was conducted with the leaves as explants. The inducing medium for callus was MS+0.2 mg/L NAA+0.5 mg/L 6-BA. The induction rate after three weeks was 100%. The most suitable medium for subculture was MS+0.02 mg/L NAA+0.1 mg/L 6-BA. The outk-tube rooting rate were relatively better without taking root in the bottle. The best matrix for planting effect was peat soil and perlite with the mixed proportion of 1∶1 and the survival rate of >95%.

非洲紫羅蘭為苦苣苔科植物,植株矮小,花型花色豐富,花期長,多季開花,是極好的室內觀賞植物。非洲紫羅蘭不易得到種子,且種子也不易萌發,因此常用葉插繁殖,但葉插殖周期長,易染菌腐爛,且其繁殖系數很低[1-2]。而組織培養的繁殖方式能更好地保留母株的遺傳特性,對非洲紫羅蘭育種的研究有著重大的意義。近年來關于非洲紫羅蘭組織培養技術的研究逐漸增多,耿明清、朱立明、紀麗麗、程云清等均以葉片進行了組培快繁技術的研究,確定各階段培養基,但對其煉苗移栽少有提及或不詳細[3-6]。本研究目的在于優化再生體系,提供適宜的煉苗移栽方法,為工廠化生產提供科學依據,也為良種選育打下基礎。
1材料與方法

非洲紫羅蘭紫色白邊單瓣花品種,取植株中部健康、完整的葉片。各……
