落葉松胚性愈傷組織誘導與未成熟胚形態的關系1)

摘要以13落葉松未成熟胚為材料進行胚性愈傷組織的誘導,其中,長73-44、日5*、日5、日5×長77-1、日5×興9、日12×興9等6個家系誘導出胚性愈傷組織,7月1日采集的日5×長77-1未成熟胚的胚性愈傷誘導率最高,達13.93%。研究結果表明,不同采集時間的落葉松合子胚形態存在一定區別,誘導出的愈傷組織形態也有所不同,其胚性愈傷組織誘導明顯受到取材時間及家系的影響。落葉松胚胎發育存在多胚現象,胚性愈傷組織的誘導率與散粉后時間及胚胎長度呈負相關,而與多胚發生率呈顯著正相關(p=0.028)。

With immature embryos for embryogenic callus induction, we induced embryogenic callus from zygotic embryos ofLarixolgensis73-44,L.kaempfer5*,L.kaempferi5,L.kaempferi5×L.olgensis77-1,L.kaempferi5×L.gmelinii9 andL.kaempferi12×L.gmelinii9. The embryogenic callus induction of immature embryos ofL.kaempferi5×L.olgensis77-1 was the highest in July 1 st, which was 13.93%. There was a certain difference in the morphology of the different acquisition time of the larch, and the callus formation was different and the callus induction was significantly influenced by the time and the family. Polyembryonic phenomenon was in the development of larch embryos. The embryogenic callus induction rate was negatively correlated with loose powder after days and embryo length, and was a significant positively correlated with polyembryonic occurrence rate (P=0.028).

落葉松(Larixssp.)屬于松科(Pinaceae)中的落葉松亞科(Laricoideae)落葉松屬(Larixmill),具有分布廣、抗逆性強及早期速生等優點,為北半球溫帶山區與寒帶氣候條件下重要的速生用材樹種,具有很高的生態價值與經濟價值。但是落葉松生長周期長,扦插等無性繁殖存在生根率較低等問題,且落葉松具有雜合度高的特點,有性生殖產生的子代存在變異,很多優良性狀難以維持[1],通過傳統的繁育方法并不能很好的滿足良種生產的需求,所以利用現代生物技術手段,建立穩定且高效的落葉松再生體系如體胚發生體系對于落葉松優良種質的繁育有積極意義,而胚性愈傷組織的誘導是體細胞胚發生體系建立的關鍵環節?!?br>
