摘要:采用熒光定量PCR方法分析了豬DIO3基因在組織中的表達狀況。結果表明,DIO3基因主要表達于背部脂肪組織,在肝臟、子宮、腎臟、心臟、小腸、肌肉和胃組織中呈中度表達,在脾臟和肺臟中表達量很低。采用外顯子687 bp處的A/C突變(A/C 687)檢測該基因在豬胚胎組織中的印記狀態,DIO3基因在所有組織中均表現為印記。

Abstract: Real-time RT-PCR was performed to analyze the porcine DIO3 gene tissue expression pattern. The porcine DIO3 gene was expressed predominantly in backfat, mildly in liver, uterus, kidney, heart, small intestine, muscle and stomach, and almost absent in spleen and lung by realtime fluorescence quantitative PCR. A single nucleotide polymorphism in exon (A/C687) was used to investigate imprinting status in porcine embryonic tissues. The results indicate that DIO3 was imprinted in all the tested tissues.

甲狀腺素脫碘酶Ⅲ(Deiodinase iodothyronine type Ⅲ,DIO3)能夠使四碘甲狀腺原氨酸(T4)和三碘甲狀腺原氨酸(T3)內環脫碘導致甲狀腺激素失活[1],在甲狀腺激素代謝過程中起著關鍵作用[2]。甲狀腺激素是調節機體生長發育及調控碳水化合物、脂肪、蛋白質等三大營養物質代謝的重要因子,DIO3基因通過調節甲狀腺激素的代謝而影響機體生長發育及營養物質的代謝,因此,DIO3是調控生長發育和營養代謝的重要調節因子。該基因在成人組織中活性很低,且僅表達于皮膚、子宮和腦等組織[3],但是在蛻膜組織、胎盤組織和胎兒組織中表達量較高[4,5]。
基因組印記是一種基因表達依賴于其親本來源的表觀遺傳現象。盡管雙親來源的兩條染色體具有相同的遺傳序列,但是印記基因只表達一個親本來源的等位基因[6]。DIO3基因位于Dlk1-Dio3印記簇最遠端,在DLK1基因下游-800 kb處[7]。在小鼠中DIO3基因表現為母本印記、父本表達[8,9]。本研究采用PCR-RFLP方法分析了豬DIO3基因的印記狀態,并研究其表達模式,為進一步研究豬印記基因對豬生長發育的調控和功能基因的分子遺傳機理具有重要的意義。……
