摘要:通過單因素試驗和正交試驗,優(yōu)化微波輻射法提取金蓮花(Trollius chinensis)中綠原酸的最佳工藝,并測定金蓮花中綠原酸的含量。結(jié)果表明,最佳提取工藝條件為微波輻射功率450 W,料液比1∶25,乙醇體積分?jǐn)?shù)70%,微波輻射時間45 min,提取溶劑的pH為7。人工種植金蓮花綠原酸含量較高,為6.686%。說明人工種植的金蓮花能夠滿足藥用。

hlorogenic Acid in Globeflower and Its Content Comparison

Abstract: Single factor tests and orthogonal experiment were adopted to study the optimization of extraction of chlorogenic acid in globeflower(Trollius chinensis) by the microwave extraction method and the content of chlorogenic acid in globeflower was determined. The results showed that the optimum extraction conditions were microwave power 450 W, the ratio of the material to liquid 1∶25, volume fraction of ethanol 70%, microwave irradiation time 45 min and solvent pH 7. The content of chlorogenic acid in artificial planting globeflower was higher than the wild varieties,reaching 6.686%. The artificial planting globeflower there fore could be used in the medicinal application.

金蓮花(Trollius chinensis)為毛茛科(Ranunculaceae)植物的干燥花,又稱旱荷、寒荷、陸地蓮、旱地蓮、金梅草、金疙瘩、旱金蓮,其味苦、性寒、無毒,可治口瘡喉腫、浮熱牙宣、耳疼目痛[1]。全世界約有25種,其中我國有16種[2],主要分布于河北、山西、內(nèi)蒙古、新疆、四川、云南、陜西、甘肅、青海等省,山西、河北省是傳統(tǒng)的金蓮花道地產(chǎn)地和栽培地[3]。現(xiàn)代藥理研究表明,金蓮花具有抗菌、抗病毒等活性,用于治療慢性扁桃體炎、感冒、發(fā)燒、急性鼓膜炎、尿路感染等其他炎癥[4]。金蓮花除了主要含有黃酮類化合物外,還含有綠原酸[5], 綠原酸具有抗菌作用。因此,綠原酸含量的高低也是評判金蓮花質(zhì)量的標(biāo)準(zhǔn)之一[6]。
本試驗采用單因素試驗和正交試驗,優(yōu)化微波輻射提取金蓮花中綠原酸的最佳工藝。通過測量寧夏回族自治區(qū)人工種植金蓮花不同部位綠原酸的含量,并與野生金蓮花中綠原酸含量進(jìn)行比較來判斷其藥用價值,為生產(chǎn)優(yōu)質(zhì)金蓮花提供參考。
1 材料與方法
1.1 材料
人工種植金蓮花,采自寧夏隆德縣西北中藥材有限責(zé)任公司種植基地;……
